As the important energy source of microorganisms and body of ruminal animals,there is above 70% carbohydrate in diets.It is necessary to study nutrition regulation of carbohydrate for ruminal animal.In recent years,the research of carbohydrate focus on effect of ruminal animal feed on carbohydrate; regulation of silage feed; quality of total mix ration feed; reduced methane gas emission through feed contained carbohydrate; the development and utilization on carbohydrate feed.This article reviewed the research of nutrition of nostructure carbohydrate and structure carbohydrate,the requirement of carbohydrate and the application of carbohydrate feed.

Research advance of vitamins from 2008 to 2009 has been reviewed in this article.Vitamin A can improve the immune function of ruminants and influence on the development of ruminant adipose tissues and shows a high antioxidation activity.Vitamin D3 can raise the concentration of Ca,P in plasama, and increase the density of 25-(OH)D3in the serum, which has some effects on the beef quality. Vitamin E can improve the performance of beef cattle, elevate the milk production, reduce the incidence of breeding, and stimulate the development of reproductive organs which results in the improvement of breeding performance. Niacin can not only improve the performance of dairy cattle, but also increase the concentrations of T3,T4 in serum. Supplementing folic acid and vitamin B12 can raise the product of lactose, crude protein, and total solid yield in milk. Choliner can release negative energy balance of postpartum dairy cattle.

Two thousand and four hundred Cherry valley super-M3 parent stocks at the age of forty weeks were selected and randomly allocated to five treatment groups with three replicates,four diets containing four different compound additives (0.5%,0.2%,0.2% and 0.2%) and basic diet were given to feed these ducks for 42 days in order to study the effects of 4 kinds of compound additives on performance and egg quality in Cherry-valley super-M3 parent stock. The results showed that laying rates were increased with diets of compound additives. When compared to control group,the laying rates of group 1 and group 2 were significantly increased by 1.04% (P<0.05) and 1.97% (P<0.05).The average egg weight of four trail groups were significantly lower than that of control group (P<0.05). The egg-shape index and the percentage of yolk of group 2 was higher than that of control group by 0.05 (P<0.05) and 13.33% (P<0.05). There were significant difference on egg thickness between control group and the first,second and fourth group (P<0.05) except group three. There was significant difference on haugh unit between control group and group 1,2 and 3,and no difference between group 4 and control group. The results indicated that different kinds of compound additives in the duck feed could increase the laying rate and improve egg quality of parent stock.

20 Kazakh lambs had an average weight of (24.71±2.07) kg,which were selected randomly as subject investigated with 4 months of age from Yili Natural Grassland in the North Bank of Turks River. The research surveyed and evaluated the changes of weight, feed intake, serum biochemical indicators and hormone contents from 2008 May 15 to September 23. The results indicated that the average daily weight gain of lambs from 4 to 8 months of age were 175 g±34 g, 92 g±36 g, 158 g ±46 g and 81 g±36 g, respectively. It reached two peaks that were 4 to 5 and 6 to 7 months of age and it were higher than other months of age. From 4 to 8 months of age, respective dry matter intake of 0.65 kg±0.21 kg,1.45 kg±0.32 kg,1.74 kg±0.48 kg, 1.95 kg±0.50 kg and 2.30 kg±0.45 kg significantly increased (P<0.01).Hormone contents determination indicated that the highest NPY contents in the whole period appeared in 5 months of age (39.10 pg/mL±2.60 pg/mL), then the lowest NPY contents in 7 months of age (36.87 pg/mL±1.10 pg/mL). No difference was found in CCK, PP, SS and CORT contents in different ages of grazing lambs, however, the highest contents of CCK, PP, SS and CORT appeared in 4 months of age (45.19 pg/mL±9.70 pg/mL), 8 months of age (108.53 pg/mL±19 pg/mL), 6 months of age (160.34 pg/mL±10 pg/mL) and 4 months of age (22.1 ng/mL±4 ng/mL), respectively. GAS and T contents increased linearly by age, the highest T4 contents appeared in 5 months of age (78.3 ng/mL±12.7 ng/mL) while the lowest appeared in 7 months of age (65.83 ng/mL±13 ng/mL). Regularity for changes of MTL, SEC, GH, INS and T3 were the same, increased in 4 to 5 and 6 to 7 months of age, but decreased in 5 to 6 and 7 to 8 months of age.

Megasphaera elsdenii is one of dominant bacteria in the rumen. In according to the characteristic of energy metabolism for the ruminant, that is energy supply through the volatile fatty acids such as acetate, propionate, butyrate generated by fermentation of microorganism in rumen, and the theory of ruminal fermentated regulation, which is mainly to regulate the degradation of lactate and the production of propionate. Primary substrates (lactic acid and pyruvate) were cocultured with H6 and TnH6 for 48 h, respectively, in order to observe the fermentation of ruminal fluid in vitro. Culture fluid was sampled for analysis of pH and VFA. The results showed that the lactate in the culture fluid was utilized by the strains of H6 and TnH6. Compared to strains H6, the acetate concentrations and the ratio of acetate to propionate were obviously reduced in the culture fluid of the strain TnH6, even though the concentration of propionate was slightly reduced. The results suggested that the transponed engineering bacteria TnH6 belonged to a deleted strain of acetic acid-producing gene and it could lay a foundation of developing a new micro ecosystem preparation for prevention and control of the diseases in periparturient dairy cows.

The experiment was undertaken to comparatively study muscle chemical composition of crossbred male and female lambs of Poll Dorset × Small Tail Han Sheep. Five male and female lambs,respectively chosen from the flocks raised in intensive production system,were slaughtered at age of 7-month-old. Longissimus dorsi (LD) was sampled and muscle chemical composition was determined. The results showed that there was no significant difference in the contents of dry matter,crude protein,crude fat,ash,calcium,and phosphorus,various amino acids,total essential amino acids,total non-essential amino acids,and total amino acids,fatty acids,total unsaturated fatty acids,Mg,Zn,Cu,and Mn in LD between male and female lambs(P>0.05). The content of linoleic acid in LD of male lambs,however,was increased by 20.3%(P<0.05) and cholesterol decreased by 32% (P<0.05) compared with female lambs. Iron in LD of female lambs was higher about 26% than that of males. The present study indicated that the nutritional value of Longissimus dorsi was the same between male and female lambs slaughtered at age of 7-month-old.

To compare the effects of ET-1 and S-Nitroso-N-acetyl penicillamine (SNAP), the donor of NO, on the proliferation of rat myocardial microvascular endothelial cells (RMMVECs). RMMVECs were incubated with different concentrations of SNAP and ET-1 for 12 h. The cell morphology was observed and cell proliferation ability was determined by MTT. Compared with the control group, the inhibition ratio of cell proliferation of various concentrations of SNAP were 10.74% to 18.80%(P<0.05), while for the various concentrations of ET-1, they could promote the ratio of cell proliferation significantly(P<0.05 or P<0.01), the promotion ratio were 20% to 42%. NO could inhibit the proliferation of RMMVECs in vitro, while ET-1 could promote it, which implied that the balance of NO and ET-1 played an important role in the development and treatment of diseases in vivo.

To study the effects of high-fluoirde and low-iodine DNA damage in the lung of old rats. The experimental models were set up by treating Wistar albino rats with water fluoride (100 mg/L NaF), diet from the area of low iodine (0.0855 mg/kg), or both water fluoride and low iodine respectively. The samples of lung were obtained by rats’ decapitation at 20 months old and were made into unicellular liquid respectively for SCGE. In comparison with DNA damage in the lung cells of the control group 1(58.46%±12.21%), such DNA damage in the HiF, LI, and HiF+LI groups 2, 3 and 4, was 75.55%±7.69%, 87.55%±12.89% and 99.37%±1.76%, respectively. Moreover, the proportion of grade III lung cells damage increased by 70.00% in group 2, 40.62% in group 3, and 79.31% in group 4, as compared to the control group 1. These findings indicated that excessive long-term intake of fluoride, with or without adequate I intake, was a significant risk factor for the development of lung damage.

Primers and probe for qRT-PCR were designed based on the matrix protein gene sequence of Newcastle disease virus(NDV),and T7 promoter was added to the 5’ end of the forward primer. RT-PCR was used to get the template for transcription in vitro,and the concentration and the D value of the RNA were determined. The results of linear and specific test indicated that the template showed good linearity and specificity.The correlation coefficient was 0.998 when the template was diluted to 2.95×104 to 2.95×108 copies/μL. Storing at -80 ℃,the template still worked well after 30 days. And the coefficient of variation value of 2.95×104 to 2.95×108 copies/μL standard positive template were 0.20%, 0.28%, 1.00%, 0.54% and 0.52% respectively,showing an excellent repetitiveness of the template. tative reverse transcriptase PCR

Deoxynivalenol (DON, vomitoxin) contamination of grains is a very common phenomenon in the global.The pigs are most susceptible to DON.The research studies pharmacokinetic of deoxynivalenol and clinical normal indexes of hematology and biochemistry by intragastric and intravenous(IV) administration of pure DON.The results showed that HCT had very significant difference in 12,24 h compared with pre-administration(P<0.01);HGB and HCT were lower than pre-administration,and had significant difference in 0.33,1,2 h compared with pre- administration(P<0.05);RBC and PLT had no difference compared with pre- administration (P>0.05).In intravenous group, WBC,RBC and PLT had no difference compared with pre-administration(P>0.05);WBC,RBC and PLT were lower than pre-administration,and had significant difference in 0.17,0.33 h compared with pre-administration respectively (P<0.05);HCT and HGB had very significant difference compared with pre- administration at other times(P<0.01).Hematologic indexes had no difference effects of vomitoxin by different administration methods in pigs(P>0.05).

A study was conducted to evaluate the activation of peritoneal macrophages by polysaccharides extracted from Allium Mongolicum Regel in mice. The peritoneal macrophages were cultured at different concentration of Allium Mongolicum Regel polysaccharides, the phagocytosis capability, energy metabolism level and IL-6 activity were assayed. The results showed that Allium Mongolicum Regel polysaccharides could enhance the phagocytosis capability, energy metabolism level and IL-6 activity. Except the test of IL-6 activity, 400 μg/mL Allium Mongolicum Regel polysaccharides and the blank control group were not significant (P>0.05), in the other index, the test groups were significantly higher than the blank control group (P<0.05). The peritoneal macrophages could be activated by polysaccharides extracted from Allium Mongolicum Regel in mice.

This test was aimed to establish the method of determining chlorogenic acid in Eucommia Leaves by uv spectrophotometry. Main factors influencing the extraction of chlorogenic acid from the Eucommia Leaves were studied by single factor experiments and used uv spectrophotometry determine the chlorogenic acid’quantitative.The results showed that the detective wavelength was set at 324 nm,there was a good linear relationship in the range of 2~20 μg for chlorogenic acid,r=0.9999. The mean recovery rate of chlorogenic acid was 100.646%,and RSD% was 1.445%(n=5).Therefore,this method was quick and accurate with good repeatability and could be used for the quality control of Eucommia leaf.

The method of RT-PCR was used to multiply PCR to identify and clarify the quail embryo sex form 66 to 120 hatch hours with primers of Wpkci and β-actin,then selected four embryos of female and male respectively at each period,taken β-actin for internal standard to determine the relative quantity of embryo bcl-2 mRNA,to investigate the influences of apoptotic factors bcl-2 on early embryos of quail.The results showed that:
① The bcl-2 mRNA expression of male embryos maintained lower level between 66 to 90 h of embryo stage,and it rose slowly between 96 to 102 h,the differences were not significant (P>0.05);The bcl-2 mRNA expression of male embryos declined on 108 h(P<0.05), but it rose distinctly on 114 h compare with 108 h (P<0.05),then maintained higher level on 120 h.The bcl-2 mRNA expression of male embryos were higher than female embryos from 66 to 108 h (expect 72 h),but they were lower than female embryos from 114 to 120 h of embryo stage. ② The bcl-2 mRNA expression of female embryos maintained lower level between 66 to 90 h of embryo stage; it rose distinctly on 96 h compare with 90 h(P<0.05),and declined slowly from 96 to 108 h ,the differences were no significant of them (P>0.05), then declined distinctly between 114 to 120 h compare with 108 h (P<0.05). ③ On the stage of 84 h, the bcl-2 mRNA expression of female embryos were higher than male embryos, and the difference was significant (P<0.05); The expression of bcl-2 mRNA in female embryos were higher than male embryos at 90 and 108 h of embryo stage (P<0.01). But, the bcl-2 mRNA expression of male embryos were very higher than female embryos at 120 h of embryo stage (P<0.01).To some extent,bcl-2 mRNA expression of female and male embryos in the embryonic development was sequential,indicated that the bcl-2 genes had an important effect on the development of the quail embryos.

To identifiy and analyze the three Arcanobacterium pyogenes isolates from Naemorhedus goral and Pseudois,the biochemistry test,analysis of 16S rRNA, automatic micbiology analysis and the other classical methods. The results showed that the three strains were Arcanobacterium pyogenes. The results of phylogenetic analysis showed that the homology of three strains campared with NCTC5224 (type strain) were 100%, and they were in the same branch.

To study the biological function of porcine alveolar macrophages FcγRⅢ,we cloned cDNA of FcγR Ⅲ gene from total RNA of porcine alveolar macrophages (PAM) by reverse tmnscription-polymerase chain reaction (RT-PCR) and analysised its sequence in this study. Sequence analysis showed that the porcine FcγR Ⅲ cDNA cloned was 820 bp in length which contained 771 bp open reading frame(ORF).The nucleotide sequence of porcine FcγR Ⅲ shared 99.9% homology with the published sequence(AF237453).The comparison of the deduced amino acids sequence of porcine FcγRⅢ with those of human,cattle,horse,sheep,monkey,dog,cat amd mouse showed that the amino acids homology similarity were 61.6%,62.9%,55.3%,62.2%, 63.0%,59.0%,61.8% and 53.2% respectively.The analysis of deduced amino acids sequence indicated that FcγRⅢ was a typical type I transmembrane protein with two Ig-like domains in extracellular region.The clone of porcine alveolar macrophages FcγR Ⅲ lay the foundation for further study of structure and function of porcine alveolar macrophages FcγRⅢ.

To investigate characterization of a mastitis resistance gene from dairy cow’s, tracheal antimicrobial peptide (TAP) gene from dairy cow’s mammary gland tissue was amplified by RT-PCR and then cloned into pMD19-T simple vector for sequence analysis. The result of DNA sequencing demonstrated that the cDNA sequence contained a open reading frame (ORF) of 195 bp which shared the highest identity of 93.8% with the TAP of bovine and encoded a 64 amino acid prepro-peptide which contained the β-defensin consensus sequence of six invariantly spaced cysteine residues. Cloning of the complete ORF of TAP may play an important role in the further designing recombinant β-defensins product.

A real-time quantitative PCR(qPCR)assay for the detection of African swine fever virus(ASFV)has been developed by designing the primers and TaqMan probe according to the sequence of the gene K205R. The reaction solutions and conditions were optimized by using different concentrations of the primers and Mg 2+ and different annealing temperature. The mask samples were adopted to verify its actual detective efficiency by adding the plasmid DNA into swine liver tissue. The qPCR detective method of ASFV based on p72 recommended by OIE was selected and modified to be as control method. Results showed that the amplification efficiency of the developed qPCR based on detection of K205R was better than that based on p72 and was sensitive and specific enough to detect African swine fever virus infection.

To establish a method for the determination of the content of Ivermectin and the entrapment efficiency of Ivermectin in the liposomes. Waters surfun C18 column (150 mm×2.1 mm, 5 μm) was employed, and the mobile phase consisted of acetonitrile∶methanol∶water (62∶30∶8). The detective wavelength was set at 245 nm. The flow rate was 1.0 mL/min, and the column temperature was at 30 ℃. Four kinds of methods were used for the separation of Ivermectin liposomes and free Ivermectin. There was good resolution between Ivermectin and the excipients by adopting to ultracentrifugation under this chromatographic condition. The calibration curve was good linear relation the range of 1 to 50 μg/mL. The mean recovery was 98.54%. The method was sample, accurate, sensitive and applicable for the determination of content and entrapment efficiency of Ivermctin liposomes.

In order to determine the DNA methylation profile of sheep Cdkn1c, its mRNA sequences of sheep, cattle, human and mouse were subjected to bioinformatic analysis in the present study. Then bisulfite sequencing method (BSP) was used to examine the DNA methylation pattern of CpG island in Cdkn1c. The results of bioinformatic analysis exhibited that the coding region of Cdkn1c was a big CpG island in four species. And bisulfite sequencing results showed that all the analyzed CpG sites were completely non-methylated in the tissues of kidney and lung, suggesting that the analyzed region was not differential methylated region (DMR) in sheep.

In this study, a pair of primers were designed according to the published DNA sequence of MDPV and was used to amplify the NS2 gene of MDPV by polymerase chain reaction(PCR). The PCR product was then cloned into the vector pMD18-T. In order to construct a prokaryotic expression vector containing the NS2 gene of MDPV, double digestion(BamH I and Sal I)were performed to deal with pMD18-T and pET-32a respectively for the directional ligation of the two fragments. The following restriction endonuclease analysis has proved the correctness of the expression vector. Construction of the prokaryotic expression vector has laid the foundation of the expression of NS2 protein and the development genetically engineering vaccine.

A pair of pecific primer was designed using Primer 3.0 software to amplify the sequence of KIFI gene from goat genomic DNA. The KIFI was cloned into pGEM-T vector,positive colonies were sequenced and digested by restriction endonuclease after transforming. The results showed that the sequence length was 472 bp (No.GQ988385),including Exon 1,part of 5′UTR and part of Intron 1 sequence of goat KIFI Gene,Exon 1 sequence coding for polypeptide contains 116 amino acid. Which shared 98%,90%,90%,88% and 84% homology with the sequences of sheep,cattle,horse,human and house mouse. Therefore,cloning of goat KIFI gene could be a basis for the further study on polymorphism and correlation on cashmere trait.

According to GenBank Salmonella invasion protein A (invA) gene sequences, a pair of primers were designed for amplification of specific nucleic acid fragment of 202 bp, after optimizing the PCR amplification conditions, established a Salmonella-specific, sensitive and rapid PCR detection method. Specificity test results showed that Salmonella strains are able to amplify the specific nucleic acid fragment, the results of E.coli, Pasteurella multocida, Staphylococcus aureus, Shigella, Bacillus cereus, Proteus mirabilis, Pseudomonas aeruginosa were negative. Sensitivity test results showed that the sensitivity of the method can reach 2.43×103 CFU/mL.

For exploring genetic diversity of Juema pig, researched on the polymorphism of growth hormone gene order of Juema pig, and selected sequences in 14 kinds of pig from GenBank. Full sequence of growth hormone gene of 7 Juema pigs had been cloned successfully for the first time, from which 5 Juema pigs were sequenced respectively, whose polymorphism is intron 1 contains 1 commutation and 1 transversion, intron 2 contains 2 transversions, explain that the difference between Juma pigs may be connected with controlling of intron. The variation of 155 nucleotide all were detected in coding and noncoding region by comparing Juema pig with other 14 breeds, and 119 out of them were in intron regions and 28 out of them were in exon regions, with 104 transversions, 14 commutations, 23 absences, 14 inserts among them. Among substitutive sites of basic group, most was transversion mutations; the comparision between transversion and commutations was 7.43∶1, which exists transversion bias phenomenon. The percentage and divergence number of Juema pig’s growth hormone gene homology analyse showed that homology was nearer by comparing Juema pig with other minitype pig, homology of Juema pig and other large pig was farer. Dendrogram of growth hormone gene sequence in Juema pig etc. of 15 kinds analyse, which indicated Juema pig having far distant genetic relationship with the Vize pig; Juema pig and Rongjiang pig’s genetic relationships were nearest.

Mycoplasma hyopneumoniae(Mhp) wild strain MY-99 was cultured in ATCC liquid medium,and extracted DNA. By PCR technique,the specific P46 gene was amplified. Then the P46 gene was cloned into pMD18-T vector and chose two cloned vectors sequenced. The results of sequencing demonstrated that the cloned sequence is 1134 bp,and it is coding 378 amino acids. The sequence is including three TGA codons coding Trp in 182,381 and 734 positions. Comparing the P46 sequence of Mycoplasma hyopneumoniae (Mhp) wild strain MY-99 with the sequence of standard 232 strain published in GenBank, the identity of them were 99.6% and 99.2%.

The cDNA expression library of Clonochis sinensis metacercaria was constructed for selecting stage specific diagnosis antigen gene or vaccine candidate gene. Total RNA of leucocytes was then extracted, pooled together and mRNA was isolated with a column of oligo (dT) cellulose. And cDNA was synthesized by Stratagene cDNA library protocol. The first and second strand of cDNA were synthesized using Stratagene Uni-ZAP XR cDNA synthesis kit, then ligated to EcoR I adapters and digested with Xho Ⅰ. After size fractionated with CHROMA Spin-400 column cDNA were ligated into Uni-ZAP vector and packaged in vitro to construct cDNA libraries. It was found that the size of the primary library was 9.15×105 PFU with a recombination rate of 99.5% and the titer of the amplified library was 1.6×1010 PFU/mL. cDNA library for Clonochis sinensis Metacercaria has been constructed.

Studied influences on quality of frozen semen in the process of producing rate of temperature drop for equilibration on Henan fat-tailed sheep from 100, 150, 200 and 250 mL water bath. Results indicated: ① the temperature drop time was 2.2 to 3.9 h that four kind water baths treated. The rate of temperature drop with 100 mL water baths was fastest, 250 mL water baths was slowest, 200 and 250 mL water baths was a similar in changes.② Optimum rate of temperature drop was 0.73 to 0.74 ℃/min and 0.11 to 0.12 ℃/min from 0 to 30 minute or 30 to 90 minute, respectively. ③ Optimum time of temperature drop for equilibration was 3 h. The cooling striking was least and the sperm quality was best with 250 mL water baths treated. ④ The sperm motility was not difference with 200 and 250 mL water bath treated, but it was significant difference between 250 and 200 mL treated or 150 and 100 mL treated. ⑤ The aberration rate was not difference with 200 and 250 mL water bath treated, it was significant difference between 250 and 200 mL or 150 mL treated. ⑥ The rate of acrosomal intactness was significant difference between 150 and 100 mL or 250 and 200 mL treated.

Osteopontin (OPN) is a kind of multi-functional secretory calcium-binding phosphorylated glycoprotein. In order to resolve the molecular mechanism of the formation of eggshell from the perspective of gene expression, egg-laying hens as the experimental animals in this study, using real-time quantitative PCR technique, to study the abundance expression of the osteopontin gene which was related to eggshell formation in the uterus at different time periods (2 h before egg into the uterus, 1 h before into the uterus, 5 h after into the uterus, 15 h after into the uterus, 1 h after egg production, 2 h after egg production). The results showed that 2 h before egg into the uterus, 1 h before into the uterus, 5 h after into the uterus, OPN expression levels increased gradually; 15 h after into the uterus, 1 h after egg production, 2 h after egg production OPN expression levels decreased gradually.The change of regularity was closely related to egg shell calcification.

The polymorphism of the serum transferrin was investigated in the blood samples from 74 Oura Tibetan sheep by means of PAGE. The results were as follows: five alleles including TFA,TFB,TFC,TFD and TFM were showed in total on the locus of serum transferrin in the examined Oura Tibetan sheep, and of them the TFB,TFC and TFD were the preponderant alleles, and their frequencies were 0.4122,0.2973 and 0.2162. Nine genotypes including TFAB,TFAC,TFAD,TFBB,TFBC,TFBD,TFBM,TFCC and TFDD were showed, of them TFBC,TFBB,TFBD,TFCC and TFDD were the preponderant genotypes, and their frequencies were 0.2703,0.1622,0.1622,0.1351 and 0.1216. The gene heterozygosity on the locus of serum transferrin was 0.6921.

The aims of this study were to determine the effects of VC on protection of frozen-thawed X-sperm. The different concentration of VC (0.0, 2.0, 4.0, 6.0, 8.0 mg/mL) were added in ordinary dilution for experimentize. After thawed in thermostat-controlled waterbath about 37.5 ℃ for 30 s, then to detect the changes of sperm motility and the percent of integrity perforatorium and plasma membrane integrity. The results showed that the concentration of VC as 6.0 mg/mL, the sperm motility compare with the control group indicate no differences (P<0.05) when after frozen for 0 h, and it indicated differences (P<0.05) compare withd control group when after frozen for 1,2 and 3 h.when the concentration of VC as 4.0 mg/mL have the good effect on the rate of the sperm plasma membrane integrity. It indicate differences (P<0.05)compare with the control group. When frozen for 0 hour, and indicated no difference compare with che control group (P>0.05).When the sperm frozen for 1 h, and frozen for 2,3,4 h,the volue indicate difference compare with the control (P<0.05).When the concentration of VC as 4.0 mg/mL had the good effect on the rate of the sperm acrosome integrity. It indicated differences (P<0.05)compare with the control group. When frozen for 0 h, and indicate no differenced compare with che control group (P>0.05). When the sperm frozen for 1,3 h, and frozen for 2,4 h,the volue indicated no difference compare with the control group(P>0.05),but the volue was higher than control group. These results indicated that added the sufficient VC as 4 to 6 mg/mL in dilution of frozen semen can rise X-sperm quality effectily after frozen-thawed.

Hox genes are transcriptional factor that plays an essential role in determining tissue differentiation during embryonic development. In the present study, expression pattern was detected in cashmere goat hair follicle by situ hybridization for mechanism of action of Hox genes. The results showed that positive signal of Hoxa4, Hoxa5, Hoxa6was detected in hair follicle of cashmere goat at different period, which indicated that Hox genes play a important role in development of cashmere goat hair follicle.

To discuss whether the effects of melatonin might result from nuclear receptors, mRNA was extracted from the skin of Inner Mongolia cashmere goat, and retinoid-related orphan receptor α( RORα), retinoid-related orphan receptor β(RORβ) and retinoid-related orphan receptor γ(RORγ) were amplified by RT-PCR, the expression of RORs in the skin of Inner Mongolia cashmere goat from different stages of hair follicle development was explored. The results revealed that the expression of RORα,RORβ and RORγ mRNA was detected in Jan(catagen), May(early anagen) and Sep(anagen),while non-expression in Mar(telogen),and RORα,RORβ and RORγ had the same expression pattern.It suggestted that RORs might have relationship with the cycled growth of cashmere.

Prostaglandins (PGs) are a group of lipid mediators, which were synthesized by the cyclooxygenase (COX) pathway from arachidonic acid. In mammals, it has been proved that PG plays a number of essential roles in reproductive processes, such as ovulation, fertilization, implantation, decidualization, uterus smooth muscle contraction, luteal regression and follicle development. In the hen, administration of PG induces oviposition and stimulates uterine contractility. At midsequence of ovipositions, the concentration of PG in the ovarian venous plasma was 5 to 20 times higher than in peripheral plasma.PG was released in a pulsate format in the avian, this not only stimulates uterine contractility, but also acted as the second messenger of gonadotropins for cAMP action in the granulosa cells’ proliferation. Hence,the change of PG content in follicle fluid played an important role in prehierarchical follicle selection.

This study was designed to investing the single nucleotide polymorphism and analyzing the genetic specificity of the EX-FABP gene, utilizing the PCR-SSCP technology and DNA sequencing, the 60 weeks of age and reproduction of the 12-week-old Ru-gao chicken as the experimental material. It detected three genotypes, two SNP mutations, 4271 G/A,4281 G/A respectively. The results showed that the content of AA-type was the mostly at the 60-week-old, significantly higher than the AB-type(P<0.05), using the least-squares significant difference test. At the 12- week-old, the AB-type was in the highest levels, as well as the BB-type was minimum in three genotypes. The intramuscular fat content at 60-week-old was significantly lower than that of the 12-week-old(P<0.05).

In this study,334 Tianfu goats, Boer and Chengdu Ma goats were used to analyze Pβ gene polymorphism by PCR-SSCP. Results indicated that there was one mutation site in the 5′regulatory region of FSHβ gene among three types of the goat, that was nucleobase A in place of nucleobase G in 712 site. The Chi-square test for goodness of fit showed that the frequencies of the Pβ genotypes in the population of Tianfu goats and Chengdu Ma goats agreed with Hardy-Weinberg equilibrium (P>0.05),while the Boer goat significantly deviated it (P<0.01). The results indicated that the mutation in the 5′regulatory region significantly affected the litter sizes of goat, therefore, it could be preliminarily deduced that FSHβ gene was probably a major gene or a QTL linked gene which associated with reproduction traits in goat.

The expression of nerve growth factor(NGF) mRNA and NGF protein in ovary,oviduct and uterus from luteal phase and follicular phase ovary of cattle were studied by FQ-PCR technique and Western blotting. The results showed that the mRNA and protein of NGF was present in ovary,oviduct and uterus both from luteal phase and follicular phase ovary. The amount of NGF mRNA in uterus from follicular phase ovary was significantly higher than ovary from follicular phase ovary and oviduct from luteal phase ovary(P<0.05), the expression level of NGF mRNA in other tissues was no significantly difference.

he genetic variations at M307 of FUT1 gene in foundation colony of Sutai ETEC F18+ resistant breeding pigs (total 576 individuals) were investigated by PCR-RFLP method. The relationship between FUT1 gene and some traits of growth and development were analyzed in this study. The results showed that M307 of FUT1 gene digested by Hin6Ⅰcould be divided into three kinds of genotypes(AA, AG and GG).The genotypic frequencies of FUT1 locus were 0.235(AA), 0.609(AG) and 0.156(GG) respectively, this locus deviates from Hardy-Weinberg equilibrium(P<0.01) in above foundation colony. The correlation between three genotypes and the performances showed that the growth and development of individuals with AA genotype were higher than those of individuals with AG and GG genotypes. Body length of pigs with genotype AA was significantly higher than that of AG and GG genotypes. The 35-days body weigh of individuals with AA genotype was significantly higher than those with GG genotype. Body height and rear quarters width of AA genotype were significantly higher than those with AG genotype. The results indicated that AA genotype at M307 of FUT1 gene had good genetic effects on disease resistance traits of PWD and PED, as well as some early productive performances. And such association could be applied in the genetic breeding practice in the near future.

he objective of this study was to explore the effects of buffalo follicular fluid (BuFF) and bovine follicular fluid (BoFF) on the IVF of buffalo oocytes obtained from Ovum pick-up (OPU) and abattoir,in order to develop IVF system and improve embryo production efficiency in buffalo. According to the replacement amount of fetal bovine serum (FBS) by FF in IVM medium,oocytes were distributed randomly into 4 groups. group Ⅰ: 0% FF+ 10% FBS,group Ⅱ: 5% FF +5% FBS,group Ⅲ: 10% FF+ 0% FBS and group Ⅳ: 15% FF+ 0% FBS. As for addition of BuFF,the results showed that BuFF did not improve the cleavage rate of OPU and abattoir-derived oocytes after IVF (P>0.05). However,addition of 5% and 10% BuFF have significantly improved the develop efficiency of IVF embryos,the blastocyst formation rates of groups Ⅱ and Ⅲ were significantly higher than those of groups Ⅰand Ⅳ(P<0.01),and there was no difference between groups Ⅱ and Ⅲ (P>0.05). The blastocyst formation rate of group Ⅳ was lower than that of group Ⅰ,but the difference between two groups were not significant (P>0.05). As for the addition of BoFF,the cleavage rate and blastocyst formation rate of OPU-derived oocytes in group Ⅲ were significantly higher than those of groups Ⅰand Ⅳ(P<0.01),and there was no significant difference between groupsⅠand Ⅱ(P>0.05). BoFF did not improve the cleavage rate of abattoir-derived oocytes after IVF (P>0.05). However,the blastocyst formation rate of groups Ⅱand Ⅲ were significantly higher than those of groups Ⅰ(P<0.05) and Ⅳ (P<0.01),and blastocyst formation rate of group Ⅳ was significantly lower than that of group Ⅰ(P<0.05),while the blastocyst formation rate was significantly different between groups Ⅱand Ⅲ(P>0.05).In conclusion,addition of 5% to 10% BuFF or BoFF in IVM medium in place of FBS can significantly improve the efficiency of IVF embryo production in buffalo,and BuFF shows a better effect.

his study was aimed to explore the effects of different methods for boar sperm capacitation by chlortetracycline fluorescence (CTC) staining.Depending on the fresh boar sperm,the discrepancy of their capacitation were compared among the induction by swim-up,A23187 and caffeine incubation for different durations by CTC staining.The results showed that the rate of sperm capacitation by swim-up method and incubating for 40 min,were significantly higher than the others (P<0.05).It could be concluded that the swim-up method was exactly suitable for the application of sperm capacitation for in vitro fertilization of swine.

To study the oxidative damage and the antioxidant status and mechanisms of them in cows with varying degrees endometritis. The content of nitric oxide(NO), malonaldehyde(MDA)and the activity of superoxide dismutase(SOD), glutathione peroxidase(GSH-Px) in serum and uterine secretions of endometritis cows were detected. The total white blood cells and percent of PMN in CE and SE were significantly higher than the control(P<0.01, P<0.05). The content of NO and MDA in serum and uterine secretions of clinical endometritis cows was significantly higher than the control (P<0.01),but subclinical endometritis was not different, and the activity of SOD and GSH-Px with clinical endometritis was significantly lower than control group (P<0.01,P<0.05). It could be concluded that the increasing of free radicals and lipid peroxide and the imbalance of antioxidant defense system in serum and uterine secretions of endometritis cows was one reason of cytopathological damage in uterine tissue.

The curative effect of combination spray of Dandelion against subclinical mastitis was observed. The cows with mastits by CMT were divided into three groups.The cows of group I were cured by combination spray of Dandelion.The group Ⅱ and Ⅲ were control group with Chinese herbs and blank.The cure rate was 88.9% and 56.5%,and the efficient rate might be the better curative effect against subclinical mastitis in dairy cow.

8 clinical health dairy cows (control group)and 16 foot-rotten dairy cows were selected and were collected serum, milk, urine, feces, wool and hoof horn before feeding in the morning, the feedstuff were collected, too. The all collection were dried and then measured the zinc level by atomic absorption spectrophotometer.The result showed that the zinc levels in serum and hoof horn of the foot-rotten group significantly decreased(P<0.01), in comparison with the results in control group, but no significant difference was observed in milk, urine, excrement and wool (P>0.05). The analysis of zinc levels in diet indicated that the zinc deficiency in foot-rotten cows seemed no relevance with the diet. The zinc deficiency, especially zinc deficiency in hoof horn, implied the closely relationship between the occurrence of foot rot and zinc deficiency.

The co-infection incidence of chicken anaemia virus (CIAV), reticuloendotheliosis virus (REV) are increasing in present poultry industry, in order to enhance the detection efficiency of CIAV and REV,according to the sequence of whole gene of CIAV and the LTR of REV published in GenBank, two pairs of primers were designed,the double PCR of REV, CIAV was established. Moreover,two kinds of extraction method of DNA were compared. The results showed that the object fragments were amplified and hold high sensitivity and good specificity. Results showed the establishment of double PCR assay was effective in the clinical diagnosis of co-infection of CIAV and REV.

One specific pairs of primers, which were designed by canine parvovirus(CPV) VP2 gene sequence, were used to clone the VP2 gene isolated from Beijing through PCR. VP2 gene was subcloned from pEASY-T-VP2 into prokaryotic expression vector pET-32a. Sequence analysis confirmed that the VP2 gene was inserted correctly. SDS-PAGE and Western-blotting analysis revealed that the VP2 protein was expressed at high level in Escherichia coli. Through ELISA assay, the expressed VP2 protein has fine immunogenicity.

In order to observe the protection of the compound recipe of Echinacea to IBDV, the Hailan Brown chickens were infected with IBDV artificially, and divided into 8 groups (30 chickens for each group), they are the compound recipe of Echinacea groups(which include the high dose group, the moderate dose group and the low dose group) and the following control groups: the Chinese medicine control group and the Western medicine control group, the vaccine control group, the negative control group and the healthy control group respectively. The results indicate that the compound recipe of Echinacea can decrease the damage of IBDV to the immune organ of chicken. Therefore, it has an obvious protection to chicken infected with vIBDV.

Chinese medicine Herba Hedyotis diffusa Willd. and Microsphaera linicerae Wint. in Rabenh and other heat-clearing and detoxifying drug as basis, supplementing with Astragalus membranaceus (Fisch.) Bunge and Glycyrrhiza uralensis Fisch., fluidextract is concentrated by water extraction. With Newcastle disease virus (NDV) as a model virus, the average time of death (MDT) and allantoic fluid HA on the chick embryo level were used as measurement indicators, the results indicate that Chinese herbal compound fluidextract significantly inhibited NDV no matter effected or not, extending MDT of the chicken embryo and reducing HA, which provides experimental basis for clinical medicine.

Trichophyton mentagrophytes genomic DNA was made as a template, using fungal general primers (ITS1, ITS4) PCR amplification of rDNA on the ITS segment to gel electrophoresis. The results showed that the cultivation of clinical samples had the same PCR polyacrylamide gel electrophoresis as corresponding strains. This experiment established an accurate, sensitive, relative specific and micro-volume method for determination of pathogenic fungi in molecular biology.

Compound Enrofloxacin injection has certain practical value in clinical. It can control effectively infection of the mycoplasmal pneumonia of swine(MPS) and alleviate the symptom, reduces the disease incidence rate and morality rate and make weight increase in a certain degree. The effect of Enrofloxacin and its combination with sulfadiazine sodium to MPS is more superior than only Enrofloxacin and only sulfadiazine sodium. This study used 2.5% compound Enrofloxacin to treat MPS of artificial infection by 0.08,0.04 and 0.12 mL/kg body weight. The optimal dose was 0.08 mL/kg body weight through intramuscular injection.

One strain virus(GD-09) was isolated from dead pigeon by SPF chicken embryos. This strain could agglutinate chicken erythrocytes and the heamagglutination of virus could be inhibited by Newcastle disease virus antiserum. It’s mean death time(MDT) of chicken embryos was 57.6 hours and its intracerebral pathogenicity index (ICPI) in one-day-old chicken was 1.74. That indicated it was a virulent strain. The cleavage site of the virus F protein was 112RRQRRF117,and accorded with highly pathogenic strain character. By analyzing genotype and homology of GD-09 and other NDV strains, it showed that GD-09 belonged to genotypeⅥ, and had lower homology compared with traditional strains La sota,F48E9,B1 and V4.

In order to formulate scientifically immunization programs to control PRRS basically, the maternal antibodies and immune antibodies of sows, sucking pigs and piglets were detected respectively by ELISA diagnostic kits. Trials have proved that the first immunization of the breeding sows was 15 days before hybridization, and the second immunization of pregnant sows was 2 months before pregnancy, the third immunization of the breeding sows was 15 days before the second hybridization, this production cycle of immunization can control PRRS infection. Because after the parturient sow 1 month immunized PRRS inactivated vaccine, the maternal antibody of piglets decreased gradually and was negative at 60 days, the first immuniza-tion of piglets was approximately 45-day-old, besides. When 40-day-old piglets PRRS were immunized inactivates vaccine, the antibody S/P average of 58-day-old was the highest, and the 160-day-old was negative which could not resist the infection of PRRSV. Injecting the 40-day-old piglets were immunized 2 mL PRRS inactivated vaccine by intramuscular, the second immunization was 70-day-old, then the antibody S/P value was the average maximum at 85-day-old, the 180-day-old finishing pigs was still positive. Therefore, the pigs were safe as the stock exchange, or immunized before hybridization as a back-up sows in breeding. The antibodies of non-immune sows and piglets were negative in control group. The fluctuation law of PRRSV maternal antibodies and immune antibodies by the experiment affords a theoretical basis for the development of immunization programs in the future.

According to the gene sequences in GenBank of APP apxⅣ A gene, one pairs of specific primer was designed for amplifying the specific fragments of apxⅣ A gene. After optimization of annealing temperature and primers concentrations, PCR was established for simultaneous detection of the APP. The specific bands of 600 bp was amplified. The sensitⅣity and specificity tests showed that the PCR was highly sensitⅣe in 50 CFU/mL. No band was amplified from Staphyloccocus aureus Rosenbach, Streptococcus, Pasteurella multocida, Salmonella typhimurium, Haemophilus parasuis, Escherichia coli by PCR, which could be stored over 12 months at -20 ℃. 41 clinical samples were detected by the PCR, which were coincided with their bacteriological test. The results revealed that the established PCR assay was sensitⅣe,specific and reproducible and it could be used to detect rapidly APP.

In order to investigate JEV infection rate and the serum antibodies level of vaccinated porcine in Guizhou province, 764 serum samples collected from large-scale piggeries and rural scatter-feed were detected by ELISA. The results showed that the antibody positive rate of JEV was 13.25% in non-vaccinated porcine and the antibody positive rate of JEV was 82.18% in vaccinated porcine, which indicated that the immune effect of JEV was more ideal. The porcine breeders should pay more attention to JEV, because the antibody levels were higher in non-vaccinated porcine in Guizhou province.
