Clone and Sequence Analysis of Dairy Cow’s Mastitis Resistance Gene: Tracheal Antimicrobial Peptide Gene

Abstract: To investigate characterization of a mastitis resistance gene from dairy cow’s, tracheal antimicrobial peptide (TAP) gene from dairy cow’s mammary gland tissue was amplified by RT-PCR and then cloned into pMD19-T simple vector for sequence analysis. The result of DNA sequencing demonstrated that the cDNA sequence contained a open reading frame (ORF) of 195 bp which shared the highest identity of 93.8% with the TAP of bovine and encoded a 64 amino acid prepro-peptide which contained the β-defensin consensus sequence of six invariantly spaced cysteine residues. Cloning of the complete ORF of TAP may play an important role in the further designing recombinant β-defensins product.
