Influence of Adding Virginiamycin to Diets of Steers on Ruminal Fermentation Parameter and Microbial Populations

The purpose is to investigate the influence of virginiamycin on rumen fermentation parameter and ruminal microbial populations of steers. Four ruminally cannulated steers(559.4 kg±30.1 kg) were used in a repeated cross-over design. The addition dose of virginiamycin was 30 mg/kg concentrates (DM). It was indicated that virginiamycin could suppress Amylolytic bacteria populations and Proteolytic bacteria populations (P<0.01).Virginiamycin increased ruminal pH (P<0.05) and the utilization rate of ruminal NH3-N (P<0.01). Viginiamycin inhibit the concentration of L-lactic acid (P=0.304). TVFA showed no difference (P=0.864). We presume that the virginiamycin inhibit Amylolytic bacteria populations and Proteolytic bacteria populations, which lead to the decrease of lactic acid concentration and consequently the pH value.

The experiment was conducted to study the effects of different dietary metabolism energy (ME) levels on adipose deposition and lipoprotein lipase (LPL) gene mRNA expression of Beijing-You chicken. Four treatments were designed in this experiment, 4 ME levels (12.122, 12.540, 13.376 and 12.958 MJ/kg) were included, 480 male chickens with 5 replicates of 24 birds each. The birds were fed the designed diets from 21 d to 91 d of age and with 70 days of trial period. LPLgene mRNA expression levels in abdominal fat were determined with quantitative fluorescence polymerase chain reaction. The results showed there were significant effects of dietary metabolism energy levels on all the adipose deposition index and LPL gene mRNA expression levels. The IMF content of breast and thigh muscle, SFW and SFR reached their peaks at ME3, LPL gene mRNA expression levels reached the peaks at ME2. Significant positive correlation was observed in LPL gene mRNA expression levels and AFR, SFW, SFR. The results suggest that dietary metabolism energy has significant effect on adipose deposition and LPL gene mRNA expression in abdominal fat of Beijing-You chicken. Adipose deposition of Beijing-You chicken was effected by LPL gene expression.

To explore the feasibility of beet silage ,we conduct an experiment to survey the effect on the quality of beet silage by adding the series of additives, namely, the formic acid, lactic acid bacteria, lactic acid bacteria plus formic acid and the calcium carbonate. The test was divided into five sub-groups, and in each group we operate three repetitions. In the test, the green fermentation tank was used. The period of the silage was 60 days. The result of the whole experiment showed that the quality of beet silage can be improved by adding the formic acid, lactic acid bacteria, and lactic acid bacteria plus formic acid. It also revealed that addition of the calcium carbonate can not obtain an obvious impact on the quality of beet silage. Considered the nutritional and sensory quality, the formic acid and the lactic acid bacteria can be served as the additives of beet silage, and the mixture of both can be better.

The rumen digestion trial was conducted to determine the effects of new bred sorghum forages in rumen of lactating cows on digestibility of energy. Sorghums in this experiment came from new bred variety, were respectively No.1 Tiannong sorghum forage A and No.1 Tiannong sorghum forage B, which dried by nature air with grains and straw. Two ruminant fistula lactating cows were used in this experiment. Two sorghum forages respectively was incubated and removed in rumen after 0, 2, 4, 8, 12, 24, 36, 48, 72 h, It was observed that sorghum forages resulted in energy digestion. The energy digestibility have respectively significantly difference at 0, 2, 4, 8, 12, 24, 36, 48, 72 h from sorghum forage A or sorghum forage B. The energy of sorghum forage A and sorghum forage B, were respectively 11.22 and 15.38 MJ/kg. The energy digestibility of sorghum forage A and sorghum forage B, were 59.41% and 62.69% at 72 h in rumen respectively. The planting technique of sorghums was able to affect the digestibility of energy. The physical and chemical performance of bred sorghum affected the digestion in rumen.

The method of invitro was used, the orthogonal design of L8 designed, to investigate effect of different combination of mixture isoacids that including isobutyric acid ,isovaleric acid and 2-methyl nail acid on rumen fermentation in cow and find out their appropriate ratio. The result showed that different combination of mixture isoacids had no significant influence on pH of rumen (P>0.05); the NO.1 combination of mixture isoacids could significantly decrease the concentration of NH3-N of rumen to 18.36%(P<0.05) as well as increase the concentration of BCP and digestion of the residue of organic dry matter by 13.43% and 10.97%(P<0.05). In this case, the evaluation of MFAEI of this concentration was positive. Therefore, we could conclude that the combination of 45%∶33%∶22% of mixture isoacids could release the protein degradation and increase the concentration of BCP and the digestion of organic dry matter residue.

The experiment was conducted to optimize the formula and processing of dog dietary for teeth care. The experiment was adopted orthogonal experimental design with three factors(wheat gluten,gel,glycerol).It could be concluded that hardness of dietary varied with the proportions of wheat gluten; the supplement of glycerol could influence hardness and the appearance of dietary; supplements of gel could influence hardness and toughness of dietary. The formula of dogs dietary was made up of 35% wheat gluten, 6%gelation, 15% glycerol, 15% water, 27% puff cornstarch, 0.5% food emulsifier and others1.5%. The dog food could be processed in accordance with nature of raw material. At first, wheat gluten, cornstarch, food emulsifier and starch were mixed up, then gel was taken into glycerol, and then raw material was added water to mix up and press them into a pie. At last, the pie was put into a drying oven of 72 ℃ for 6 h.

The objective of this study was to explore the changes of oxidative and antioxidative system of the lung tissue in BALB/c mice under normal condition. Malondialdehyde(MDA) concentration,the activity of xanthine oxidase(XOD) and superoxide dismutase(SOD)were measured by spectrophotometer. The results showed that the MDA level,the activity of XOD and SOD in the lung tissue of BALB/c mice did not show any significant difference on day 3, 6, 7, 12(P>0.05). The results indicated that the system of enzyme of generation and clearance about free radical in the lung tissue of BALB/c mice remained dynamic balance.

To observe the effects of efflux pump inhibitor-No.1 small peptide on antibacterial activity of antiseptic drug, the minimal inhibitory concentrations of eight kinds of antiseptic drug treated with No. 1 small peptide alone and in combination to twelve strains E. coli isolated from chickens were measured with standard microdilution method. The results showed that nine of twelve chickens E. coli were multi-drug resistant isolates, which can produce extended-spectrum β-lactometers. With the effect of No. 1 small peptide (one to two), the antibacterial activity of eight kinds of antiseptic drug, such as enrofloxacin, could be improved by twice and the antibacterial activity of Florfenicol against A8 and A15 was improved by fourth. With the effect of No. 1 small peptide (one to two or one to four), the antibacterial activity of Enrofloxacin, Gatifloxacin and Ciprofloxacin could be improved by twice and the antibacterial activity of Levofloxacin against A13 was improved by fourth. These results showed that bacterial efflux pump inhibitors-No. 1 small peptide one could improve the antibacterial activity of most antiseptic drugs and chickens E. coli which were produced enzyme and multi-drug resistanted has at least two resistance mechanisms of ESBLs and efflux pumps at the same time.

Studies on effects of selenium,copper,zinc in serum of different kinds of sheep in pregnancy which were added selenium in the identical condition of feeding management.There were 85 sheep in pregnancy including Mongolian sheep,Dorset and Suffolk sheep, determined contents of selenium by fluorescence spectrophotometry method, determined contents of copper and zinc by atomic absorption spectrophotometry method.Results showed that from pregnant 3 months on, the content of selenium in serum of each kind sheep which was added selenium was significantly higher than that of the same kind sheep which was non-added selenium, and at the same time, the content of zinc in serum was non-significantly (P>0.05) between the same kind of sheep which were added selenium and non-added selenium,and the content of copper in serum was non-significantly (P>0.05) between added selenium and non-added selenium Mongolian sheep.Results indicated that there is no antagonism between selenium and copper or selenium and zinc,so added selenium could play an important role to maintain natural pregnancy and to enhance resistibility.

In the present study, twenty of 2-crossbred York×Rongchang piglets were divided randomly into two groups, each for ten litters. Fructo oligo saccharides (FOS, 0.75%) were added into food for group Ⅰand groupⅡ as a control to indicate the effects of FOS in food to immune function in piglet. Blood were collected on 30 and 60 days of age to examine the Erythrocyte rosette forming cell rate (E-RFGR) and lymphocyte transformation rate (PHA). The results showed no significant difference on E-RFGR between group Ⅰand group Ⅱ on 30 days (P>0.05), while there were a considerably higher PHA in group Ⅰ than in group Ⅱ (P<0.05); Both E-RFGR and PHA on group Ⅰwere also higher than that of group Ⅱ (P<0.01)on 60 days. The above results showed that the food including FOS could improve the immune function in lymphocyte of piglets.

100 SD rats weighting 100~120 g were divided randomly into 3 groups. The animals in groupⅠwere fed with 3.0 mg/kg selenium as Se-enriched malt (SEM) supplemented diets. Animals in groupⅡand groupⅢreceived 0.1 mg/kg selenium in sodium selenite only and were severed as the positive control and negative control respectively. Animals groupⅠand groupⅡwere induced hepatoma by diethylnitrosamine resolved 〖JP+3〗with sterilized water (100 mg/L) at the dosage of 10 mg/kg body weight every day for 16 weeks,then drank with sterilized water for another 2 weeks. At 18th week,the rats were killed. Microvascular density (MVD) and.vascular endothelial growth factor (VEGF) protein level in tumor tissue were determined by immunohistochemistry. The results indicated that MVD and VEGF protein levels in the SEM group were significantly lower than those in the model group (P<0.05). It demonstrated that SEM could inhibit angiogenesis in the rat hepatoma. The down-regulating of VEGF expression might be involved in the angiogenesis inhibition.

n cattle rumen,the number of Streptococcus bovis and the type of fermentation have a direct effect on ruminal normal physiological activity.In the mean time, lactic acid which is produced by Streptococcus bovis’s fermentation and all kinds of enzymes which exist in Streptococcus bovis have important values in practical industrial production. In this test, author isolated strain from ruminal fluids,and used the microcrystalline biochemical event reactons and the 16S rRNA gene technology to identify the biochemistry and inherited characteristic of the isolated strain respectively, binding to the viewing result of gram staining and morphology, author assessment the isolated strain was Streptococcus bovis, providing the theory and experiment foundation for study and research the streptococcus bovis preferably.

In the experiments 50 broiles reared to 14 d are selected ,randomly is divided into 5 groups of 10 broilers.During 14 to 42 d the test group Ⅱ, Ⅲ, Ⅳ are administrated continuously ChIFN-α in drinking water followed by 0.1,1.0 and 10 U/mL respectively. Ⅰ group as the control group is only given water.Ⅴ group as positive control is administrated 50 mg/L levamisole hydrochloride. The experiments is designed to study the effects of ChIFN-α on immune function and growth performance after the broilers are administrated in drinking water. The results show ChIFN-α can increase the total number of leukocytes in peripheral blood of broilers,the percentage of lymphocytes, the percentage of basophils,reduce the percentage of neutrophils and monocytes, raise chickens spleen index,as well as the concentration of serum total protein (TP), serum albumin (ALB),serum globulin (GLO) and ALB/GLO ratio; and can not impact on the broiler growth and development obviously after drinking administration. As a result, administration of low-dose ChIFN-α in broilers drinking water can enhance the immune function of broilers, and can not impact significantly on the growth the healthy broilers.

An indirect enzyme-linked immunosorbent assay (ELISA) method was established to detect antibodies against porcine circovirus type 2 using the recombinant capsid protein (rCap) expressed by prokaryotic pQE30 vector as coating antigen. The various factors affected the experiments such as concentration and coating time of rCap, the serum dilution concentration, etc were optimized, and the storage period of the ELISA kit was also tested. The results demonstrated that optimal concentration of rCap for coating was 2.815 μg/mL, the optimal coating condition of rCap for ELISA was incubated at 37 ℃ for 2 hours, the dilution of serum sample was 1∶40, the work concentration of HRP-labeled rabbit anti-porcine IgG was 1∶500, the best blocking buffer was BAS with 1% concentration, and the critical value of positive and negative of ELISA was D450 nm=0.33. The specificity test showed that the established ELISA method was only displayed D450 nm>0.33 for standard positive serum of PCV-2, but D450 nm<0.33 for other serums of PCV-1, CSFV, JEV, PRV, PRRSV, etc which acting as the negative controls. Sixty-nine of 100 clinic suspicious PCV-2 infection samples was positive by using the established ELISA method. The storage period test of the ELISA kit showed that the kit could be stable for at least 12 months at 4 ℃. This study indicated that an indirect ELISA method for detection PCV-2 in serum was successfully established, and could be used for the rapid clinic diagnosis of PCV-2.

A pair of primers was designed based on the h gene sequence of canine distemper virus in GeneBank, the h gene of (CDV) strain isolated from an attenuated vaccine strain of canine distemper virus was amplified by reverse transcription polymerase chain reaction (RT-PCR). The amplified fragment was cloned into pMD18-T simple vector and the positive recombinants were identified by blue-white screening and restriction endonuclease digestion, and the h fragments were sequenced and analyzed. Sequencing analysis indicates that the gene was the same to the representive canine distemper virus h gene. h gene was 90.6%,91.3%,90.5%,91% identical to Yangzhou isolates,Changchun isolate,XinJiang isolate and Japanese isolateD85755 respectively. It was 97.7%,97.2%,98.1% indentical to Onderstepoort strain,Convac strain and American isolates 98-2666-2 respectively. Then the h gene was subcloned into Prokaryotic expression plasmid pET-32a(+). Recombinantplasmid carrying h gene (pET-32a-h) was transformed into E.coli BL21(DE3) and induced with IPTG. A fusion protein about 64 ku was expressed.

he VP1 gene of FMDV type Asia I was inserted into the prokaryotic expression vector pET-28a, and then transformed into BL21 cells. The recombinant plasimd was induced with different concentration and time of IPTG to express the VP1 protein. The better expression condition was confirmed by analyzing the effect of different concentration and time of IPTG on the expression product. The experimental result indicated that the expression product had not been increased with adding of the concentration of IPTG in definite scope, but the expression output had been increased with the extension of time during the period of logarithmic growth of bacteria. The maximal quantity of expression of VP1 protein was confirmed when the concentration of IPTG was 3 mmol/L and the induced time was 9 hours.

In order to study the role of chicken toll like receptor 4 playing in Salmonella infection, eighty three-day-old specific-pathogen-free chickens were randomly divided into 2 groups. The treatment group chickens were orally inoculated with 0.5 mL of the bacterial suspension containing 108 CFU/mL Salmonella Serovar Pullorum. The control groups were inoculated with 0.5 mL saline. Eight chickens of each group were randomly chosen and killed at day 1, 3, 7 and 14 post infection. Spleen was sampled for RNA extraction. SYBR GreenⅠquantitative real-time PCR method was applied to compare the relative expression quantity of chicken TLR4 gene. The following results were obtained: TLR4 gene expression levels were significantly higher in chickens from treatment group than those in the control group at 4 time points (P<0.05), especially at day 3 and 7 post infection (P<0.01). These results indicated that the expression of chicken TLR4 gene was up-regulated during Salmonella infection.

Karyotyped and chromosome banding of Guishan goat and Red-bone goat was analyzed by the techniques of peripheral blood lymphocyte culture and the techniques of making chromosome specimens. The result indicated that the karyotype of two has rarely no diffrents, number of goats chromosome is 2n = 60. Among them, 29 pairs are autosome and 1 pair is allosome, X chromosome is that for 2nd pair of big telomeric ends of chromosomes, Y chromosome is the smallest and the only centromeric chromosomes. The phenomenon of chromosomal numerical change were finded both in Guishan goat and Red-bone goat.The results of C-banding and G-banding is shown that there is no significant different between two breeds.

The preference of serum is very significant during the cell culture. The experiment used the commercial serum, self-made newborn bovine serum and self-made adult bovine serum to culture mechanocyte, myeloma cell and hybridoma cell and then applied the MTT to verify the cell growth better or worse, so that we can evaluate the quality of the serum. The results demonstrated that self-made newborn bovine serum was the best for cell growth (the relative growth rate of every above cells exceed 0.96), the three experimental data showed the result was not different (P>0.05), so the self-made newborn bovine serum was more stable. However the self-made adult bovine serum and no serum comparison had not the significant effect to promote the cell growth. So the MTT could estimate the quality of the serum.

Transfer vector is considered important step in the construction of recombinant virus, based on the sequence analysis of the genome of goatpox virus, based on the KpnⅠof the pUC119-TK, transfer vector with the P7.5 controlled EGFP was constructed. After transfection in BHK21 cells infected with goatpox virus, the reporter gene expressed 48 hours later. The results lay foundation for recombinant vaccine development.

A pair of primers was designed to clone the POU1F1 cDNA according to published swine POU1F1 mRNA sequence. The total RNA was extracted from swine pituitary and the full length cDNA of swine POU1F1 was amplified by reverse transcription-polymerase chain reaction (RT-PCR). The PCR product with expected size of 876 bp was inserted into PTZ57R/T vector and sequenced. Finally the result sequence was analyzed by DNAStar and it demonstrated 99.7% homology with published sequence in GenBank. The cloned cDNA will be used for further function research of alternative splicing of swine POU1F1 Gene .

Two complementary sense and antisense oligo-nucleotides, encoded bovine lactoferricin B gene (LfcinB), were synthesized based on amino acids sequence of bovine lactoferricin B published in the antimicrobial peptide database (APD). After the sense and antisense oligo-nucleotides annealed, a cohesive BamH I site at 5′terminus and a cohesive Xho I site at 3′terminus were formed. Synthesized LfcinB gene and eukaryotic expression vector pCDNA3.1 (+) digested with BamH I and Xho I restriction endonuclease were ligated, and ligation products were transformed into E.coli DH5α. Plasmid extracted from transformed bacteria were sequenced by Sanger dideoxymethod. Sequencing result indicated the synthesized LfcinB gene was successfully cloned to multiple cloning site of vector pCDNA3.1 (+). This may lay a foundation for expression and antimicrobial activities in future.

The diseases of sheep and goat pox and lumpy skin disease of cattle are caused by members of the capripoxvirus. Capripoxvirus, the member of the orthopoxvirus family with the largest genome, probably encodes more genes involved in cell adaptation than any of the other members.Recombinant virus vector is one of the most hot spots in virus gene engineering, there are two vector type, the replaceable recombinant virus vector and the recombinant viral plasmid vector. This review summarized recent advances in the methods of construction of viral transfer plasmid, homologous recombinant, and the activated vaccines derived from the viruses deleted the nonessential gene or inserted with neutralizing gene of other pathogens.

A total of 372 pigs from Duroc, Landrace, Large White, Mashen, Shanxi lean-type SD-Ⅰline, SD-Ⅱ line, SD-Ⅲ line and Shanxi white pig were used to detect the polymorphism within intron 8 of the prolactin receptor gene (PRLR) by PCR-SSCP. Three alleles of A, B, C and four genotypes of AA, AB, AC, and CC were detected in the examined populations. The frequencies of allele A were higher in all examined breeds (lines), ranging from 0.50 to 0.62, than those of allele B, with the range of 0.12 to 0.44. The frequencies of allele C were quite low, ranging from zero to 0.04 in the examined pig populations with the exception of in Duroc (0.26) and in SD-Ⅲ line (0.26). Three transition mutations of G→A,T→C,A→G and insertion mutation of TTT were detected by cloning, sequencing and homologous comparison of the homozygote AA and CC. The records of litter size at first parity of 140 sows and the records of growth and development of 118 replacements of Shanxi white pig were used to investigate the association of PRLR genotype with pig performance by using least square analysis. The results showed that no significant differences both in TNB and NBA at first parity were found among the different PRLR genotype sows. However, the significant associations between PRLR genotype with total teat number, body weight and backfat thickness at the age of six months were detected. The total teat number of individuals with AA genotype was significant higher than those of AC, and the body weight and backfat thickness at six-month age of the replacements with AC genotype was significant higher than those of replacements with AB genotype (P<0.05).

Ghrelin gene and pituitary specific transcription factor 1 (Pit-1) gene were studied as candidate genes on the Chaohu duck in this study. Polymorphisms of Ghrelin and Pit-1 gene had been analyzed by PCR-SSCP and DNA sequencing method. The results showed that three SNPs were identified at Ghrelin gene, one SNP (G54A) located at the third exon, another two in the fifth exon were transitions of C to A (149) and G to T (166). The frequency of A/B alleles for Ghrelin gene in the population was 0.49 and 0.51, and that of M/N alleles was 0.19 and 0.81. No polymorphisms were detected in the amplified regions of two pairs of primers in Chaohu duck population tested, which may indicate that the sequences of exons 3 and 4 of Pit-1 gene were rather conserved.

In this study,mRNA isolated from embryos was subjected to reverse transcription polymerase chain reaction to investigate the expression of eNOS, iNOS mRNA in normal pathenogenetic activation porcine embryos. The results showed that eNOS was expressed only in two-cell and four-cell stage embryos, while iNOS was expressed around early embryo development stages. These studies determined that inducible NOS (iNOS) and endothelial NOS (eNOS) are differentially distributed in cells along the development, higher level of eNOS was detected at four-cell stage embryo but iNOS at morula/blastocyst stage. The study provides the first evidence for the presence of mRNA of NOS isoforms during porcine embryogenesis. Furthermore, the expression of NOS during the embryo development suggest that NO may be of crucial importance for reproductive events that occur in this period.

The Mx gene in Anka chicken was analyzed with mismatch PCR-RFLP method, the results showed that M2032 locus of Mx gene digested by restriction endonuclease RsaⅠcould be divided into three kinds of genotypes, AA, GG and AG. The frequencies of these genotypes were 0.232, 0.681 and 0.087, respectively. The frequency of resistance allele A was 0.572. The result of Chi Square test indicated that the frequencies of Mx gene digested by RsaⅠ were significantly deviated from Hardy-Weinberg equilibrium in this study. The correlation between the three genotypes with some commercial traits was analyzed, the results showed that the body weight of different developmental stage, body size mainly slaughter traits and meat quality of the individuals were with resistant genotype AA were not significant different with those individuals with AG and GG genotypes in Anka chicken, which indicated that M2032 locus of Mx gene had no significantly negative genetic effects on commercial traits in Anka chicken.

The genetic diversity of mitochondrial DNA D-loop 5′-terminal partial sequences of 378 individuals in eight Chinese goat (Capra hiruc) breeds were analysised by PCR-SSCP technology. The results show that seven haplotypes (A, B, C, D, E, F and G) with the percentage of 1.9% in 378 animals were obtained in eight populations. The haplotype diversity was higher as 0.8003 for the Taihang black goat, and lower as 0.5772 for the Hongtong Milking goat, The results showed that the genetic diversity of Chinese goats is abundant.

Hair follicles were isolated from goat ear skin by 2.4 U/mL Dispase digestion and the bulges were separated with special fine needle and digested with 0.5 mg/mL trypsin plus 0.2 mg/mL EDTA. The results showed that cells were cultured in different concentrations of IGF-1, IGF-1 had a good effect on the goat hair follicle stem cells in vitro and the 10 ng/mL was best concentration.

The syndrome of porcine reproductive and respiratory syndrome (PRRS) is characterized by respiratory disease leading to increased mortality in piglets and decreased performance infertility or late-term abortions in sows. The change of pathology and microanatomy was not often typical in infected pig, it was easy to confused with other pathological changes caused by other etiological agents, it is difficult to diagnose. The definite diagnosis was depended on laboratory diagnosis to increase the level of diagnosis and prevention. The sensitivity, specificity and cost are different in different laboratory diagnostic assays,serology test can’t differentiate antibodies caused by field virus or vaccine. The research about methods of viral RNA detection, viral antigens, and antibodies in this article were discussed.

Two kinds of inactivated oil-emulsion pseudorabies virus vaccine were used to inoculate rabbits which were not detected PRV antibody,every group had three rabbits,the control group was not inoculate vaccine.Their serum were collected at regular intervals.Latex agglutination test(LAT) and neutralization test(NT) were used to detect the antibody in serum .The results showed that LAT method can detect PRV antibody after 7 days ,and NT method can detect PRV antibody after 21 days.After 28 days inoculated,rabbits were infected with PRV.The experimental groups did not show significant clinical symptom, the control group rabbits were all died. The results showed that when the PRV LAT≥1∶45.3 or NT≥1∶8 ,the rabbit inoculated with the vaccine could resist PRV infection.

A bacterial which was Gram positive, slenderness and irregularity in appearance were isolated from a pig, its liver had purulence nodus and lungs and the spleen were haematocele. The isolated bacterium grew slowly in BSA, sheep’s blood agar and chocolate agar, and showed α-haemolysis in sheep’s blood agar. The results of 16S rDNA PCR sequence and API biochemistry identification showed that the isolated bacterium was Arcanobacterium pyogenes. The Arcanobacterium pyogenes caused 100% mouse death in 24~52 h, and cause rabbit death in 72 h by enterocoelia innoculation, it showed that it was virulent bacterium.

In order to investigate the prevalence of porcine reproductive failure caused by viruses, 1068 patient samples from 14 pig farms in the vicinity of Taiyuan were detected using multiplex PCR diagnostic methods. The average infection, rate of porcine reproductive and respiratory syndrome virus (PRRSV), classical swine fever virus (CSFV), porcine parvovirus (PPV), pseudorabies virus(PRV) and porcine circovirus type 2 (PCV-2) were 32%,40%,20%,12%,27%, respectively.More than two viruses mixed infection was 39%,two virus infection was 24%. 1572 blood samples were detected using RT-PCR kits for PRRSV RNA and CSFV RNA in 11 cities of Shanxi province from 66 pig farms and 121 free-range pig farm households. The results showed that PRRSV infection rate was 33%. CSFV infection rate was 46%.1593 blood samples were detected to indentify immune antibody titer of CSFV, the average qualified rate was 43.9%. It provides experimental data for working out comprehensive measures to prevent and control the epidemic diseases.

Effect of four commonly used immunization program of Newcastle disease in commercial broiler were detected under the high maternal antibody titer, the results indicated that the level of maternal antibody had decreased significantly, the change was not significant in group but significant between groups. After the second inoculation, maternal antibody still had great interference with the live vaccine group, the antibody titer below the protection level(6 log2) at 55 days old, but the antibody titer of the 7-day-old group higher than 3-day-old group. Maternal antibody had small interference with the group used both inactivated vaccine and live vaccine, antibody level of 3-day-old group up to 6log2 at 50 days old while antibody level of 7-day-old group up to 6.4log2 at 45 days old and rose to 7log2 at 50 days old and maintained to the end of trial. The antibody level of 7-day-old group which was about five days ahead of schedule to achieve the level of antibody protection was higher than the 3-day-old group.

In order to investigate the effects of erzhu fuling san and its components on the tension of isolated caecum in chook, isolated intestine was adopted, the chook’s caecum was put in the magnus’ bath (maintained (41±0.5) ℃ constant temperature), then the vary dosage decoction of rhizoma atractylodis, atractylodes macrocephala, tuckahoe and erzhu fuling san were added to observe the effect on the tension of isolated intestine. The result indicated that erzhu fuling san and its components (the decoction of atractylodes macrocephala, rhizoma atractylodis and tuckahoe) restrained the movement of intestine, reduced the contract and diastole tension and contract swing of chook’s isolated caecum.The effects of all kinds of the decoction enhanced with increase of the concentration.

The investigation of parasite infection was taken in 22 kind of herbivores,and 88 feces samples were collected in Zhengzhou zoo. The centrifugation and saturation sugar solution floatation were used in survey.The result showed that the gross infection rate was 19.13%,and the rate of coccidian infection,nematode infection and whipworm infection were 7.95%,12.50%,2.27%, respectively. The analysis of data showed that the rates of parasites infection were decreased gradually,and degrade rate was 39.95% compared with 2003,and the extent of infection reduced obviously. It also showed that the protection measure in the zoo had a good effect,all above offered reference and basis for the zoo to make correlative measure on the prevention of parasites disease in the future.

To determine the prevalence of intestinal parasites in pet chinchillas, the sheather’s sugar flotation, modified acid-fast staining and Lugol’s iodine-solution staining methods were used to examine the faecal samples of chinchillas in a pet market in Zhengzhou city. A total of 96 fresh faecal specimens were examined and two protozoan species (Giardia spp., coccidia) and one helminth species (Hymenolepis nana) were found. The overall infection rate was 46.9%. Giardia spp. had the highest prevalence (37.5%), coccidia (8.3%) and Hymenolepis nana (2.1%) took the second and third place. No statistical significance was found between the infection rate of Giardia and the age (P>0.05) and gender (P>0.05). This is the first report of intestinal parasites in pet chinchillas in China. 

The objective of this study was to estimate genetic parameter for locomotion score by applying single-trait animal model through MTDFREML with REML algorithm in China Holstein cattle, and data from 887 cows collected in 2007 derived from Wuhu farm in Wuhan. The results showed that locomotion score was affected by lactation number and housing types. There was an increasing tendency in locomotion score with the increase of lactation number. Locomotion score of tie-stall dairy cow was higher than that of free-stall dairy cow, the heritability of locomotion score was 0.09, the locomotion score, was a low heritability trait.

A goose paramyxovirus isolated from dead goose in goose farms of Chongqing was determined by the methods of serology and biology. Objective to study the infectious condition of goose paramyxovirus and Newcastle disease virus in the area, the sera of 124 chickens, 59 ducks and 121 geese were detected by HI test, respectively. The results showed that the goose paramyxovirus and Newcastle disease virus positive infection rates of the chicken were 70.1% and 83.9%, which of the duck were 1.7% and 6.8%, while of the goose were 61.2% and 55.4%. It indicated that the infectious condition of goose paramyxovirus was serious in the area, and there was cross immunogenic properties between the two antigens.

To study the pathological examination effect of ZhenTian capsule, the rats were orally given ZhenTian capsule for 180 days with the doses of 1.315, 2.63 and 5.26 g (crude medicinal materials)/kg (13, 26.1 and 52.2 times of clinic does) while the control group was given the distilled water of the same volume and for 4 weeks of stopping administration. Meanwhile, rats’ behavior and appearance were observed, and night shit and urine everyday amounts were measured, coefficient of the main organs and pathological examination of the rats were investigated. All the examinations and index are within the normal level. No significantly histological change related to visceral is observed.

In broiler-fryer industry of Chongqing, there has a disease which harmed 15 to 30 days old broilers since September 2007. The disease occurred as an acute septicemia characterized by fibrinous perricarditis, perihhepatitia, and airsacculitis. In order to search for the etiopathogenisis and the means of cure, we isolated and identified the pathogenic bacterium. In the study, SS nutrient agar was used, and 26 strains Salmonella enteritidis (SE) were isolated and identified by biochemistry and PCR. The SE strains had pathogenicity for chicken, ducklings and mice. Drug susceptibility test for 26 strains showed their resistance to Amoxicillin, Ofloxacin, and so on. The chicken immunized with CQ0709 strain inactivated vaccine or injected with special IgY can against artificial challenge. The results showed drug resistance SE infection had been a problem in the intensive production of broiler in Chongqing.

Apramycin was diluted to different concentrations and was added to bacterium suspension which is made of Staphylococcus aureus and antibiotics standardization medium Ⅲ. After cultivated for 3 to 4 h at 37 ℃, the linearity of the dose-response relationship was analysised. The accuracy of developing turbidimetric method was checked. At the concentration range from 2.9 to 7.2 U/mL, log values of concentrations of apramycin were linear correlation with corresponding opacities, which correlation coefficient was r=0.9988. By the t-test, regression effect of log values of concentrations of apramycin with corresponding opacities was significant. Average recovery is 99.82%, and average confidence rate is 2.48%, and relative standard deviation is 1.86%. Potencies were unitied which determined by turbidimetric method and cylinder-plate method. Turbidimetric method has low confidence rate, high sensitivity, high precision,so that can be used to determine apramycin and apramycin peprations.
