Abstract: The preference of serum is very significant during the cell culture. The experiment used the commercial serum, self-made newborn bovine serum and self-made adult bovine serum to culture mechanocyte, myeloma cell and hybridoma cell and then applied the MTT to verify the cell growth better or worse, so that we can evaluate the quality of the serum. The results demonstrated that self-made newborn bovine serum was the best for cell growth (the relative growth rate of every above cells exceed 0.96)， the three experimental data showed the result was not different (P>0.05), so the self-made newborn bovine serum was more stable. However the self-made adult bovine serum and no serum comparison had not the significant effect to promote the cell growth. So the MTT could estimate the quality of the serum.
