Identification of 3 Exons Polymorphisms in the Endothelial Nitric Oxide Synthase Gene of Chinese Merino Sheep by PCR-SSCP

Abstract: This experiment was to study the single nucleotide polymorphisms (SNPs) and single amino acid polymorphism (SAPs) of exon 7,exon 13 and exon 14 in Chinese Merino sheep endothelial nitric oxide synthase (eNOS) gene, which were analyzed according to the published SNPs of exons loci of endothelial nitric oxide gene of human and mouse in GenBank. 3 exons of Chinese Merino sheep eNOS gene, having the higher similarity with the human and mouse, were found through bioinformantics method, and they were exon 7,exon 13 and exon 14, respectively. Exons polymorphism in human and mouse eNOS gene were aligned and analyzed by the NCBI SNP database, found that 2 SNPs loci in the human eNOS gene exon 7 (rs1799983) and exon13 (rs148554851) had clinical significance. After that, the amplified fragments of the 3 exons in Chinese Merino sheep eNOS gene were analyzed by PCR-SSCP method, indicating that SNPs loci were not discovered 3 exons in Chinese Merino sheep, therefore, 3 exons regions of Chinese Merino sheep eNOS gene were relatively conservative.
