锰致鸡肾脏蛋白质氧化损伤作用的研究

Study of Manganese on Protein Oxidative Damage of Kidney in Chicken

摘要: 为探讨锰致鸡肾脏组织蛋白质的氧化损伤作用及其分子机制,分别在日粮中添加500、800、1700 mg/kg MnCl2染毒处理30、60、90 d,建立鸡亚慢性锰中毒模型。采用2,4-二硝基苯肼比色法测定鸡肾脏组织蛋白质的羰基含量,用以判断蛋白质的氧化损伤程度。结果表明,与对照组相比,各染毒组肾脏组织中的蛋白质羰基含量增大,且呈一定的时间-剂量效应。因此,锰能使鸡肾脏组织蛋白质羰基含量增加,从而诱导机体氧化损伤。

Abstract: In order to explore the oxidative damage effect and the molecule mechanism of manganese on kidney protein of chicken. 500, 800, 1700 mg/kg MnCl2 were added to the forage to establish the model of the sub-chronic manganese poisoning. 2,4-dinitropheny hydrazine was used to detect the protein carbonyl content to evaluate the degree of protein oxidative damage. The results showed that the protein carbonyl content of kidney increased in each poisoned group compared with control group, and there was a time-dose relationship. Therefore, it is demonstrated that manganese could increase the protein carbonyl content, and induce oxidative damage.
