Detection of CLPG and MSTN Genes Genotypes on AWF Sheep

Abstract: Callipyge (CLPG) and myostatin (MSTN) are recognized as the growth of muscle-related genes in the world,which produce "beautiful buttocks " and "double-muscle" good traits after mutation. To study whether imported AWF sheep carrying mutations in two genes,we used PCR-RFLP technique to check. The results showed that there was a C→T SNP in CLPG gene,AWF sheep populations had CC and CT genotypes;A G→A SNP locus in the 3'-UTR region of MSTN gene, AWF sheep populations had GG and GA genotypes. Analysis of variance with the R language between different genotypes and growth in different gender,MSTN and CLPG of different genotypes on the AWF rams weight,height,length,rump width,fat thickness and loin area were no significant differences (P>0.05);CLPG CT genotype on ewe weight was significantly higher than CC genotype (P<0.05),in length was very significantly higher than CC genotype (P<0.01),MSTN GA genotype on ewe loin area was very significantly higher than GG genotype (P<0.01). We detected contain CLPG and MSTN mutations in imported AWF sheep from Australia,and the mutation of sheep body traits had significant effects.Therefore,the AWF sheep could used as improved local sheep breeds,gene pyramiding and terminals supporting male in ternary system after the introduction.
