Analysis on Polymorphism of KRT26 Gene and its Association with the Wool Fineness in Fine-wool Sheep

Abstract: The present study was designed to investigate the traits of economic importance of molecular genetic features of functional genes, to evaluate the genetic relationship in fine-wool sheep, to provide molecular genetic basis for efficient breeding of sheep breeds, and to effeciently conserve and utilize the germplasm resources.PCR-SSCP, DNA sequencing and bioinformatics techniques were used to analyze the association of KRT26 with the wool fineness in 289 fine-wool sheep.The results showed that in fine-wool sheep population, there were three genotypes and the genotypic frequencies of AA, AB, BB were 0.221, 0.426 and 0.353, respectively.The gene frequencies of allele A and B were 0.434 and 0.566, respectively, which was intermediate polymorphism with the polymorphic information content of 0.371 and was not in the agreement with Hardy-Weinberg equilibrium (P< 0.05).Applying BioEdit and Chromas software, five novel SNPs of KRT26 gene were genetyped, which were 83 bp(G/C), 86 bp (T/C), 112 bp (C/T), 140 bp (G/A) and 247 bp (C/T), respectively.Moreover, two novel SNPs were missense which resulted in change in amino acid (SNP 140G/A lead to Val/Ile and SNP 247C/T lead to Asn/Lys).The association of SNPs with wool fineness trait showed that the wool fineness with AA genotype were extremely significant higher than that with AB and BB genotypes (P<0.01).The results implied that the KRT26 gene might be one of important candidate molecular markers associated with the wool fineness trait.
