Quantification methods of rumen microbes mainly include molecular and traditional methods which comprise classical roll-tube technique and most probable number technique. Many studies proved the results of numbering microbes with traditional method were possiblely lower. However, quantification of important groups or species using molecular-based procedures can help explain their role in ruminal metabolism but also to provide perspective for more qualitative approaches. This paper reviewed quantification methods development phases of rumen microbes and recent studies, focusing on quantitative aspects of those using molecular biology approaches.

Objective： to study the effect of admixture of cordyceps culture medium with different percentage on blood biochemical parameter in SPF Wistar rats. Methods： we collected eighteen items blood biochemical parameters of sixty SPF Wistar rats divided into three groups, and then these parameters were run statistical analysis. Results： Levels of HG, EOS of Wistar rats in experimental groups are lower significantly than control group's; Levels of CH, TGL of Wistar male rats in experimental groups are lower remarkably than control group's, while Levels of UN, BG of its are higher notably than control group's; Levels of GPT, GOT of Wistar female rats in experimental groups are higher significantly than control group's, while Levels of CH, TGL of its are lower notably than control group's; other parameters among other groups and different sexes do not exist difference remarkably. Conclusion： part blood biochemical parameters in SPF Wistar rats generate significant change by feeding admixture of cordyceps culture medium with different percentage.

In order to discuss the influence of sodium humic acid added on feed for goat to the use of mineral element,60 adult guanzhong dairy goats were randomly divided into two groups,30 in each group.In test group,4 g sodium humic acid was provided for each goat every day.The ration of control group was not changed during test period.At the end of the feeding experiment,the mineral elements in the milk and dejection from the two groups were estimated.The result showed that the levels of P、Fe、Cu、Zn in milk from test group was lower than that of control group,the levels of Cu and Zn were only 53.57% and 38.07% as more as that of control group.The level of Ca、P、Fe、Cu、Zn in dejection from test group were 1.32,1.23,1.49,1.28 and 1.68 times as that of control group respectively.Thus it can be seen that the mineral elements was lost excessively and their utilization ratio was decreased by the odium humic acid added to feed for goats.

in the blood was 2.28 mmol/L, the milch cow behad myasthenia of limbs. While it got to the concentration of 3.45 mmol/L after the therapy, the milch cow could get up itself. During the whole diagnosis, we partly adopted the therapy to diminish inflammation, reduce gastric acid secretion and stop bleeding by astringency.

In this experiment, allantoic and amniotic fluid volume, pH value were mensurated and analysed. The results showed that amniotic fluid content reached the peak value at the 16th day, and its pH value staied at 7.0 all the time. Allantoic fluid had the highest content at the 12 th day, and its pH value was more than 7.0 all the time, which also achieved the peak value at the 12th day. The PH value of amniotic and allantoic fluid supplemented each other to some degree.

The primary cultured cells of horse skin tissue were obtained by explant tissue culture and were purified by enzymes digest. Firstly, Horse skin fibroblasts were frozen in 12 different freezing protective media, which contained different concentration of DMSO, EG, GC and NCS. Secondly, Horse skin fibroblasts were frozen in two different cryo-preservation methods. Efficiency of attachment at 24 h was used to evaluate the effects of cryopreservation. The results of experiment showed that DMEM supplemented with 10% DMSO and 20% NCS was the most efficient cryo-preserving media (efficiency of attachment at 24 h after thawing, 84.98%). The effects of cryopreservation in programming-cryo-preservation were obviously better than those in manual cryo-preservation (86.32%, 79.98%)(P<0.05).

In this experiment, the effect of cytochalasin B (CB) on vitrification of ovine GV-stage oocytes were analyzed. This experiment included toxicity test,vitrification and pretreatment with different cytochalasin B (CB) concentration. The results were as follows: rates of matured oocytes of toxicity groups both, with or without CB pretreatment were lower than control group (P<0.05), but there was no difference between them(P>0.05). In vitrified groups, the rate of matured oocytes was 16.1% and 14.1% respectively after in vitro maturation culture, they were all significantly lower than control group (P<0.05). 9.0ug/ml of CB could improve the maturation rate compared with control group(P<0.05).

The new collected semen removed the seminal fluid was mixed with the linearized plasmid DNA pro data, then stay it at 17℃for 2h let the DNA got into the sperm head. Sperm cells needed to penetrate and fertilize eggs by artificial insemination, the 2 pregnant gilts generated 20 piglets, and four positive transgenic pigs were detected by PCR. The research results proved that the human her2 gene had integrated into the pig chromosome, the integration rate was 20%.

This paper focused on isolated primordial germ cells (PGCs) from gonads at stage 19 by Ficoll densitygradient centrifugation. Then, the PGCs were froze respectively in six kinds of freezing medias under slow speed cryopreservation process and vitrification process. The viability of the frozenthawed PGCs was measured by the trypan blue exclusion method. The results showed: For the slow speed cryopreservation, the viability of the frozenthawed PGCs under freezing media V was the highest, and showed significant difference（P<0.05）when compared to that of freezing mediaⅠ.For the vitrificated cryopreservation, the viability of the frozenthawed PGCs under freezing media I was the highest,and showed very significant difference（P<0.01）when compared to that of other five freezing medias. The third passage PGCs subcultured after slow speed cryopreservation and the second passage PGCs subcultured after vitrificated cryopreservation were positive to PAS and AKP staining and still kept intact karyotypes.

RNA interference(RNAi) is a post-transcriptional gene silencing induced by introduction of double-stranded RNA (dsRNA)。RNAi is achieved through incising by nucleic acid into small inference RNA of 21~23 nt by dsRNA, which is called siRNA, and is recognized and targeting incises the homologous mRNA molecular. In order to detect the polymorphism of heart fatty acid-binding protein (HFABP)in the SongLiao black pig , genetic variations of 5’-upstream region and the second intron in 62 SongLiao black pigs H-FABP gene were checked by PCRRFLP with HinfI, HaeIII and MspI.The results indicate that SongLiao black pig only shows sigle AA genotype in MspI-RFLP, for HinfI-RFLP, SongLiao black pig had polymorphism and frequencies of allele H is 0.7177, and for HaeIII-RFLP, SongLiao black pig had polymorphism and frequencies of allele d is 0.6210.So the experiment indicates that SongLiao black pig has polymorphism in the 5’-upstream region and the second intron of H-FABP gene.

According to the IGF-Ⅰgene 5’-UTR conserved sequences of chicken, duck etc, a pair of degenerate primer was designed to amplify the sequence of Wulong Goose IGF-Ⅰgene 5’-UTR from the genomic DNA. And then the PCR product was subcloned to the pMD18-T vector and sequenced which was 796 bp in length. Compared with the duck and chicken, Wulong Goose IGF-Ⅰgene 5’-UTR sequence shared 98.1％and 93.0％nucleotide homology respectively which indicated that IGF-Ⅰgenes were highly conservative in phylogeny. Moreover, it has 4 bases deletion and 7 bases insertion compared to duck, and 4 bases deletion and 3 bases insertion compared to chicken. There are 5 kind of common restriction sites through nuclear acid DNA sequence by macrorestriction map analyzing.

Karyotype analysis was carried out on Henan Game chicken using the peripheral lymphocyte culture and air drying technique. The result showed that the diploid chromosomal number of Henan Game chicken was 78,according to the measured relative length ,arm ratio and centromeric index,the first 10 pairs of chomosome were described as followed: No.1,Z chromosome were metacentrics,No.2,No.7 and W chromosomes were submetacentrics, No.3, No.6, No.8, No.9 and No.10 chromosome were telocentrics, No.4 chromosom was elocentric chromasome. Study on Henan Game chicken's G-bands showed that the first 10 pairs of macrochromosomes could be devided into 25 zones and 152 bands.

The method of saturated brine floating and McMaster`s was used in this test. In order to know the digestive tract parasite infection of carnivorous animal and omnivore in Zhengzhou Zoo, the lynx etc 11 species, 48 carnivorous animals and omnivores were investigated, and then those positive animals were treated. The result indicated that the infection rate of parasite of 48 animals was as high as 52％, but most animals' intensity were slight except one male lynx. Mebendazole was used to feed the positive animal at the dosage of 5mg/kg·BW with food at one time. No eggs was detected in feces after 5 days treatment, this show the medicine has good effect to nematode in lynx’s.

Selected 15 SAF1、DAF1 and SAF2、DAF2 which were ablactaed at 2.5 month old under the same condition to fat for sixty days, respectively. The result indicated that the average daily gain of SAF1、DAF1 was 269 grams and 262 grams, relatively increasing by 24.54% and 21.30% compared with ALT，respectively and the difference is remarkable（P<0.01）；The average daily gain of SAF2、DAF2 was 258grams and 254 grams, relatively increasing by 19.44% and 17.59% compared with ALT，respectively and the difference is better（P<0.05）；The differences of the average daily gain among SAF1、TAF1、SAF2、TAF2 are not better .During fattening period, the pure relatively increasing economic income of each hybrid lamb are 30.42 yuan、32.95 yuan、28.12 yuan、30.21 yuan.

The capability of heat-insulating and economy of the surroundings of pig houses of light steel-arched frame with polystyrene insulation were analyzed . The results showed:1)the heat resistance of wall was a little small that condensation would happen in cold winter;2)the property of roof insulation was not good because the maximum temperature of inner surface might be upper than the maximum air temperature outside in summer; 3)the fairly economic thickness of polystyrene of the roof was 50mm or so.

In the experiment about 6 monthold cross bred F1 of Simmental×Guan ling yellow cattle, Limousine×Guan ling yellow cattle and Angus×Guan ling yellow cattle were ten heads all allocated into three groups for 360 days of finishing trial. The results show that: the ADG of the three kinds of cattle is 1413 g,1485 g,1367 g respectively; the dry matter intake per 1 kg ADG is 6.91 kg,6.42 kg,7.10 kg respectively; the cost of feed per 1 kg ADG is ￥5.23, ￥4.86, ￥5.38 respectively; there are no significant difference in Net meat ratio among three kinds of cattle, the weight among 18 to 24 month about 450 kg to 500 kg .Above results indicate that straight fatting can support satisfactory performance in both growing and finishing.
