Exon 2 SNP Detection of PRL Gene and its Associations with Egg Production Traits in Wenshang Barred Chicken

Abstract: In this study, Wenshang Barred chickens was selected as the research object,exon 2 of prolactin (PRL) gene were detected by PCR-SSCP and the DNA fragments were sequenced. The correlation analysis between the polymorphism of the exon 2 and egg production traits were carried out respectively. The results indicated that there were two polymorphic sites in exon 2. C→T mutation at the position of the 3838 bp was found, which resulted in 3 genotypes (AA, AB and BB). Furthermore, an amino acid changed from leucine to phenylalanine. The frequencies of genotype BB and allele B were the highest. Three genotypes of AA, AB and BB in age at first egg, living weight at first egg, egg weight of first egg, living weight at age of 300 days, egg weight at age of 300 days index of reproductive traits difference did not reach the significant level (P>0.05). The results showed that PRL gene exon 2 polymorphism and reproductive traits of Wenshang Barred chickens presented no significant correlation.
