Designing and Screening of Microsatellite Primers in Chinese Hamster

Abstract: The objectives of the present study were to screen new microsatellite loci in Chinese hamster to develop genetic markers for genetic analysis. A library of partial small size fractionated genomic DNA was constructed with the Chinese hamster.135 pairs of primers were designed with the software Primer 3.0 for microsatellites positive clones obtained. 11 pairs primers that the stutter bands easily in theory were synthesized and 20 samples were tested with them. The results showed 10 of the 11 loci were found to be polymorphic,and PIC,the mean observed heterozygosities (Ho),the mean expected heterozygosities(He)were 0.4195, 0.3895 and 0.4565, respectively. The microsatellite markers would be useful for further studying on accessions identification and breeding of Chinese hamster,which would provide some evaluable tools for marker-assisted selection breeding and gene mapping in Chinese hamster.
