Study on Anti-inflammatory Activity of Total Flavonoids from Viola yedoensis in Vitro

Abstract: To study the effects of total flavonoids from Viola yedoensis(TFV) on the content of inflammatory mediators and the expression of related genes in LPS-induced RAW264.7 cells.Screening of the optimum concentration of TFV for promoting RAW264.7 macrophage viability by MTT method.Then its effects on the release of NO,tumor necrosis factor α(TNF-α),IL-1β,IL-6,TNF-α,iNOS and COX-2 expression on RAW264.7 cells were investigated,thus to study and analyze the in vitro anti-inflammatory activity of TFV.Results showed that TFV could increase RAW264.7 cell viability in the concentration range of 5-50 μg/mL (P<0.05).Comparison with LPS model group,TFV could significantly reduce the production of NO,TNF-α,IL-6 and IL-1β (P<0.05),and could significantly reduce the expression of TNF-α,COX-2 and other inflammatory factors in RAW264.7 cells induced by LPS.TFV could down-regulate the expression of inflammatory factors such as IL-1β,IL-6 and TNF-α,and down-regulate the expression of TNF-α and COX-2,which may be one of the reason for the anti-inflammatory effect of Viola yedoensis.
