不同抗生素组合对瘤胃厌氧真菌滚管培养过程中细菌生长的影响

Effect of Different Antibiotic Combinations on Bacteria Growth with a RollTube Technique to Isolate Anaerobic Fungi from Rumen Fluids

摘要: 摘要:作者应用亨氏厌氧滚管培养技术,结合菌落计数, 探讨了青霉素、氯霉素、链霉素和甲硝唑不同抗生素组合添加对瘤胃厌氧真菌培养过程中细菌生长和繁殖的抑制作用。固体培养基和液体培养基培养试验结果表明,单一添加青霉素和链霉素组合,并不能有效抑制细菌生长。当在培养基中将0.8~1.2 mg/mL青霉素、1.2~1.8 mg/mL链霉素与0.004~0.008 mg/mL甲硝唑进行三者组合,或将0.8~1.2 mg/mL青霉素、0.6~1.2 mg/mL链霉素、0.1~0.3 mg/mL氯霉素与0.004~0.008 mg/mL甲硝唑进行四者组合时,瘤胃细菌生长被显著抑制,而真菌生长则不受影响(P<0.05)。总之,瘤胃细菌对抗生素具有一定的抗性,本研究结果为今后有效控制亨氏滚管法分离培养厌氧真菌过程中的细菌生长和繁殖提供了抗生素浓度组合和添加浓度参考依据。

Abstract: Abstract: The objective was to study effect of different antibiotic combinations on growth of rumen bacteria with a Hungate’s roll tube technique to isolate anaerobic fungi from rumen fluids of dairy cows. Penicillin, Streptomycin, Chloramphenicol and Metronidazole in different levels were added to 9.0 mL solid or liquid mediums prior to the anaerobic inoculation. Based on counting results of bacteria and fungi up to three day, the combination of 0.8 mg/mL Penicillin and 1.2 mg/mL Streptomycin did not inhibit the growth of rumen bacteria in mediums. However, the combination consisted of 0.8 to 1.2 mg/mL Penicillin, 1.2 to 1.8 mg/mL Streptomycin and 0.004 to 0.08 mg/mL Metronidazole significantly inhibited the growth of rumen bacteria without negative impact on that of anaerobic fungi (P<0.05). Similar inhibition also occurred for the combination consisted of 0.8 to 1.2 mg/mL Penicillin, 0.6 to 1.2 mg/mL Streptomycin, 0.1 to 0.3 mg/mL Chloramphenicol and 0.004 to 0.008 mg/mL Metronidazole. In general, antibiotic resistance of rumen bacteria should be paid more attention, and the combination of different antibiotics in this study might provide an alternative reference for the effective control of bacteria growth in the application of a roll tube technique to isolate anaerobic fungi from the rumen fluids. 
