PCR-SSCP Analysis on AHCY Gene in Holstein Cows

Abstract: The AHCY (S-adenosylhomocysteine hydrolase) gene was studied as a candidate gene for mastitis resistance in 210 Holstein cows. Single nucleotide polymorphisms (SNPs) of all exons (exons 1 to 10) of AHCY gene were detected with thirteen pairs of primers by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) in 210 Holstein cows. No polymorphism was detected in the amplified region in the ten exons tested, this result indicated that the genetic markers that affect the mastitis resistance in Holstein cows may not exist in the detected exons of AHCY gene.
