香附酮抑制鸡致病性大肠杆菌诱导肺泡Ⅱ型上皮细胞炎症作用的研究

Inhibitory Effect of Cyperone on Inflammation of Chicken Alveolar Epithelial Type Ⅱ Cells Induced by APEC

摘要: 本试验通过建立鸡致病性大肠杆菌O78(APEC-O78)感染的鸡肺泡Ⅱ型上皮细胞的体外炎症模型,研究了不同浓度香附酮对TNF-α、IL-10分泌及TLR-4和TLR-5 mRNA表达水平的影响。结果显示,0.5 μg/mL香附酮能显著降低TNF-α的分泌水平(P<0.05),同时1 μg/mL香附酮能极显著促进IL-10的分泌(P<0.01)。0.1、0.5和1 μg/mL香附酮均能极显著降低TLR-4 mRNA的表达水平(P<0.01),而不影响TLR-5 mRNA的表达水平(P>0.05)。结果表明,香附酮可能通过TLR-4受体信号通路,抑制APEC-O78感染的鸡肺泡Ⅱ型上皮细胞促炎因子的产生,增加抑炎因子的产生,进而发挥其抗炎作用。

Abstract: The effects of cyperone on chicken alveolar epithelial type Ⅱ cells infected by APEC-O78 were evaluated through determining TNF-α, IL-10, TLR-4 and TLR-5 in vitro. The results showed that 0.5 μg/mL cyperone significantly decreased the secretion of TNF-α(P<0.05). 1 μg/mL cyperone extremly significantly increased IL-10 secretion(P<0.01). Furthermore, 0.1, 0.5 and 1 μg/mL cyperones all extremly significantly decreased the expression of TLR-4 mRNA(P<0.01), but had no effect on the expression of TLR-5 mRNA(P>0.05). The results indicated that cyperone might inhibit the production of inflammatory cytokines of chicken alveolar epithelial type Ⅱ cells induced by APEC-O78 through blocking TLR-4 signaling pathway.
