中药王不留行增乳活性单体及催乳素对奶牛乳腺上皮细胞特异性miRNA的影响

Influence on the Specific microRNAs in Bovine Mammary Gland Epithelial Cells by Semen Vaccariae Active Monomer and Prolactin

摘要: 本试验旨在研究中药王不留行增乳活性单体邻苯二甲酸二丁酯及催乳素对奶牛泌乳中期乳腺上皮细胞miRNAs表达的影响;荧光定量RT-PCR检测增乳活性单体邻苯二甲酸二丁酯及催乳素作用后乳腺上皮细胞miRNAs表达变化;王不留行增乳活性单体邻苯二甲酸二丁酯及催乳素均抑制原代培养的泌乳中期奶牛乳腺上皮细胞miRNA-143、miRNA-125和miRNA-195表达;邻苯二甲酸二丁酯可抑制miRNA-21表达,催乳素对miRNA-21表达的影响尚不确定。首次阐明中药王不留行增乳活性单体邻苯二甲酸二丁酯和催乳素能引起乳腺上皮细胞miRNAs表达变化。

Abstract: The objective of this research is to find the influence on specific microRNAs in bovine mammary gland epithelial cells by Semen vaccariae active isomer and prolactin. Quantitative real-time PCR was used to detect the expression changes of microRNAs (21, 125b, 143 and 195) in the mammary gland epithelial cells with dibutyl phthalate (DBP) and prolactin (PRL) at different time. The results showed that both DBP and PRL could repress the expression of miRNA-125b, miRNA-143 and miRNA-195 in mammary gland epithelial cell; DBP could repress the expression of miRNA-21,while the influence of PRL on miRNA-21 was not certain. It is the first time to demonstrate that the expression of the microRNAs can be changed by Semen vaccariae active isomer (DBP)and prolactin.
