Phylogenetic Relationship of Bos grunniens in Qinghai Based on Mitochondrial cox1 and nad1 Genes

Abstract: The objectives of the present study were to amplify and analyse the mitochondrial cytochrome c oxidase subunit 1 (cox1) and NADH dehydrogenase subunit 1(nad1) genes among Bos grunniens (B.grunniens)isolates from Qinghai province in China, and to study its phylogenetic relationships with Bos of the other species using cox1 and nad1 genesss sequences. The partial cox1 (pcox1) and nad1 (pnad1) genes were amplified from each B.grunniens sample, and pcox1 and pnad1 gene sequences were aligned using the ClustalX 1.81. Maximum parsimony (MP), maximum likelihood (ML) trees were constructed using the software Phylip 3.67 version 4.0 and ML tree was also constructed using Puzzle 5.2. The lengths of all the pcox1 and pnad1 gene sequences were 823 and 837 bp, respectively. Phylogenetic analyses showed that the B.grunniens samples from Qinghai were clustered in the same clade. The results of this study provided foundation for further studies on molecular genetics and variation of B.grunniens.
