Sequencing and Phylogenetic Analysis of Mitochondrial cox1 and nad1 Genes for Cysticercus Cellulosae in Xichang

Abstract: The complete sequences of mitochondrial cytochrome C oxidase subunitⅠ(cox1) gene and the partial sequences of NADH dehydrogenase subunit 1 (nad1) gene of three cysticercus cellulosae samples isolated from Xichang were amplified by PCR, and sequenced to analyze their genetic variations, NJ trees were reconstructed using the Mega version 5.0 software.The results showed that the complete sequence of cox1 gene and partial sequence of nad1 gene of cysticercus cellulosae were 1 620 and 796 bp, respectively.There were some genetic variations in cox1 and nad1 genes within cysticercus cellulosae, the level of variation was higher in the cox1 gene than in the nad1 gene.The phylogenetic trees showed that all the cysticercus cellulosae isolates were formed a single cluster, and the three Xichang isolates belonged to Asian genotype.The results indicated that cox1 and nad1 genes could be used in molecular classification of Taenia solium, and established the foundation for molecular diagnosis of taeniasis/cysticercosis.
