PCR Amplification and Sequence Analysis of Mitochondrial Cytochrome c Oxidase Subunit 1 (cox1) of Syphacia obvelata from Mice

Abstract: The partial sequences of mitochondrion cox1 (pcox1) gene of 5 pinworm samples separated from mice were amplified by PCR and the positive products were sequenced. Then sequences were analysed by BLAST and compared with Syphacia obvelata and S. montana available in GenBank. The results revealed that the length of the pcox1 sequence was 373 bp. Sequence alignments showed that genetic variation within 5 pinworm samples was very small, with only one mutation locus, and these samples were identified as S. obvelata by BLAST. The inter-specific sequence difference was 5.24%-5.28% between S. obvelata samples and S. montana available in GenBank. These findings showed that the intra-specific variations of cox1 were low for S. obvelata and the inter-specific sequence differences were comparatively higher between S. obvelata and S. montana. The results provided foundation for further studies of population genetics of pinworms. 
