Differential Analysis of Transcriptome in the Ovaries Between the Highly and Low Inbred Langshan Chickens

Abstract: In order to explore the effect of inbreeding on reproductive effects,this study selected Langshan chickens conserved in the National Chicken Genetic Resources as materials.Combining molecular markers and pedigre information,two groups of highly and low inbred chickens were established,and the reproductive performance data were recorded for both groups.Three chickens was selected as a material in each group,and ovarian tissues were taken for RNA-seq sequencing,and the functional annotation of different genes was carried out.The results showed that a total of 1 114 differential transcripts were obtained in the highly and low inbred groups,of which 783 genes were annotated,307 were up-regulated and 476 were down-regulated.GO and KEGG analysis showed that the differential genes were mainly enriched in biological processes such as ribosome biosynthesis,inflammatory response,reproduction,growth,immune system processes,and metabolic processes.Biological pathways including in folic acid biosynthesis,oocytes maturation and metabolism were also enriched by the differential genes.Functional analysis found that the differential genes (such as GGH,CPEB1,GNMT and PIWIL) were related to reproductive function.In addition,some genes related to immunity and stress (such as APOC3,HSP70,CD38 and LGMN) were also included.The results of this study were helpful for us to understand the genes and their regulatory mechanisms related to inbreeding depression of reproductive traits in Langshan chickens,and would provide a reference for the molecular mechanism of inbreeding depression of specific traits in poultry.
