Establishment and Preliminary Studies on the Skin Tissue Hair Follicles Proteins of 2-DE Map Building in Fine-wool Sheep

Abstract: In use of hair follicles in skin tissue of fine-wool sheep with different fiber diameters as experimental material, and different extraction methods, IPG strips with different pH and different sample volume as experimental methods, this paper was aimed to explore two-dimensional electrophoresis system applied to skin tissue of the fine-wool sheep and establish 2-DE gel map of skin tissue in fine-wool sheep.Gel maps using ImageMaster 2D Platinum software automatically detected protein points to compare different protein fiber diameter of fine-wool sheep differences.The results showed that the TCA/acetone extraction method was most suited to fine-wool sheep hair follicle tissue extraction of total protein.Choose to 18 cm, pH 4 to 7 linear IPG strip, we got better quality two-dimensional gel electrophoresis, got 35 protein variations among the different cantons for the follow-up work fine-wool sheep with high quality of the study of proteomics.
