3种抗凋亡试剂在猪精液低温保存中的应用

摘要: 为了改善猪精液低温保存方法,研究在低温条件下葛根素、酒石酸美托洛尔和二氯乙酸钠3种抗凋亡试剂对保持猪精液品质的作用,采用手握法采集健康公猪精子活力大于0.7的精液,用添加不同浓度葛根素(0、0.02、0.10、0.50、2.50 mmol/L)、酒石酸美托洛尔(0、0.02、0.10、0.50、2.50 mmol/L)及二氯乙酸钠(0、0.04、0.20、1.00、5.00 mmol/L)的稀释液稀释,于5 ℃下保存,每天检查,以精子稀释后活力、有效存活时间、总存活时间、存活指数及保存72 h后精子畸形率和顶体完整率为衡量指标,试验重复6次。结果表明,一定浓度的3种药物能有效地延长低温保存时猪精子的存活时间,其中0.50、0.50及1.00 mmol/L分别是葛根素、酒石酸美托洛尔和二氯乙酸钠的最佳添加浓度。

Abstract: The objective of the present study was to improve the low temperature preservation of boar semen and evaluate the effect of three anti-apoptotic drugs (puerarin, metoprolol tartaric and dichloroacetate) on keeping the quality of boar semen. Semen was collected from adult boars of proven fertility with the gloved-hand technique, and only the semen showing a minimum of 70% motile was used. Semen was diluted in extenders containing anti-apoptotic drugs (0,0.02,0.10,0.50 and 2.50 mmol/L puerarin, 0,0.02,0.10,0.50 and 2.50 mmol/L metoprolol tartaric acid, 0,0.04,0.20,1.00 and 5.00 mmol/L dichloroacetate). All sperm suspensions were stored at 5 ℃ and sperm motility was detected once a day. Six parameters (motility of extended soermatozoa,spermatozoal-survival effective time,spermatozoal-survival overall time, spermatozoal-survival index, the 72 h deformity rate and acrosomal integrity) were measured, six replicates were performed. The results showed that under the conditions of this study, a certain concentration of three drugs can extend the life-span of spermatozoa under the low temperature, and the optimal concentration of puerarin, metoprolol tartaric acid and dichloroacetate was 0.50, 0.50 and 1.00 mmol/L, respectively.
