摘要: 变性梯度凝胶电泳(denaturing gradient gel electrophoresis,DGGE)技术是一种新型的用于检测核酸变异和点突变的电泳方法,主要根据突变型和野生型核酸序列的不同而导致其变性浓度的差异,从而利用变性梯度胶进行分离,可用于肠道菌群结构多样性和种群动态变化的研究。文章对DGGE技术的原理、在肠道菌群多样性和动态性的应用、局限性及发展前景及上综述。

Abstract: Denaturing gradient gel electrophoresis (DDGE) was a new electrophoresis method for testing the nucleic acid variation and point mutations, mainly based on the different denaturing concentrations dued to the different nucleic acid sequence of mutant type and wild type, thus using the denaturing gradient glue to make it separate. It could be used in the diversity and dynamics analyses of microbial communities. The principles of DGGE, applications in diversity and dynamics of intestinal bacterial community, the limitations and future developing perspectives were summarized in this paper.
