梅花鹿鹿茸总蛋白提取方法对双向电泳图谱的影响

Methods of Antler Total Protein Extraction on the Effect of Map Quality of Two-dimensional Electrophoresis in Sika Deer

摘要: 鹿茸是唯一可以重复再生的哺乳动物器官。利用不同的提取方法制备梅花鹿鹿茸总蛋白样品,分别进行双向电泳,从得到的蛋白点的数量和点的圆滑程度、分辨率等确定适合的提取方法。分别用全蛋白提取试剂盒、自制裂解液、丙酮沉淀制备蛋白样品,并对得到的3种2-DE图谱进行比较分析。结果显示,试剂盒提取法和自制裂解液法得到的蛋白点比较多、圆滑,图谱清晰;丙酮沉淀法得到的蛋白点不如前者多,图谱有横竖纹。

Abstract: Deer antlers are the only mammalian organs capable of repeated regeneration. To prepare total protein samples of sika deer antler for 2-DE by different extraction methods, and then find a suitable one from the spots number and resolution condition in the map. Total protein extraction kit, manual lysate and acetone precipitation were used and three kinds of 2-DE maps were analyzed. The results showed that spots number and resolution of kit and manual lysate were better than that of acetone precipitation, and stripes in the map of acetone precipitation were visible.
