藏绵羊肌肉抑制素C-端重组蛋白抑制肌肉生长功能的研究

Study on the Function of Tibetan Sheep Myostatin C-domain Recombinant on Inhibiting Muscle Growth

摘要: 本试验旨在检测注射藏绵羊肌肉抑制素C-端重组蛋白后对小鼠的影响,研究肌肉抑制素的作用,为今后生产实践提供参考。本试验对50只小白鼠(雌、雄鼠各25只)进行分组,随机将雌雄鼠分为2组,其中对照组注射生理盐水,试验组注射肌肉抑制素蛋白,每隔1 d称重,试验期共3个月。试验期结束后,处死小鼠,并取小腿肌肉样本,利用石蜡切片法和浸渍离析法,测定小鼠肌纤维直径。试验结果表明,藏绵羊肌肉抑制素C-端重组蛋白对小鼠肌肉生长早期(37~45日龄)具有抑制作用。当小鼠体内对藏绵羊肌肉抑制素C-端重组蛋白产生抗体后,对小鼠肌肉生长的抑制作用逐渐减弱。藏绵羊肌肉抑制素C-端重组蛋白可使小鼠腿肌肌纤维横切面积增加、腿肌肌纤维直径增加。

Abstract: To observe the effect of the mice which were injected by MSTN genetically engineered proteins for studying the function of the MSTN. Fifty mice(25 female and 25 male rats) were divided into two groups, experimental group and control group both included male and femle rats. The control groups were injected normal saline(NS) and the experimental groups were injected MSTN proteins. Weighing respectively every other day for 3 months. The mice were executed after the test period, we collected their hamstrings, through paraffin section and dipping segregation methods, observed the mice’s muscle fiber diameter. Results indicated that the recombinant Tibetan sheep myostatin C-domain can inhibit muscle growth in the early time of mice(about 37 to 45 days old). When the antibodies to be produced in the blood of mice, the function of recombinant Tibetan sheep myostatin C-domain gradually be weakened. The recombinant Tibetan sheep myostatin C-domain can increase muscle fiber cross sectional area and muscle fiber diameter of mice.
