猪源A组轮状病毒Vp4基因工程乳酸菌电转化条件的优化

Optimization of Electroporation Processing of Porcine Rotavirus Group A Vp4 Genetic Engineering Lactobacillus

摘要: 为了提高pW425et-Vp4基因工程乳酸菌的转化效率,对影响电转化效率的主要因素进行研究,以得到最佳的电转化条件。试验结果表明,猪轮状病毒Vp4基因工程乳酸菌电转化的最佳条件为:当感受态细胞的D600 nm值为0.4时,在MRS培养基中加入1%甘氨酸+0.3 mol/L蔗糖,电场强度为11 kV/cm,脉冲时间为3.8 ms,电击后细胞复苏3~4 h等条件时,得到较高的转化效率,最高可达3.8×104 CFU/μg DNA。

Abstract: In order to obtain higher electrotransformation efficiency of pW425et-Vp4 engineering Lactobacillus, studied and researched the major factors which effected the electrotransformation efficiency. The results showed the optimal values for the D600 nmof competent cell, concentration of glycine and of sucrose, the electrical field strength, the pulse time and the length of revival were 0.4, 1% (W/V), 0.3 mol/L, 11 kV/cm, 3.8 ms and 3 to 4 h, respectively, best electroporation efficiency was3.8×104 CFU/μg DNA.
