高致病性猪繁殖与呼吸综合征病毒JL株 Nsp10基因生物信息学分析

Bioinformatics Prediction of the Nsp10 Gene of Highly Pathogenic PRRSV JL Strain

摘要: 本研究旨在对高致病性猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)贵州JL株Nsp10基因(GenBank登录号:EU886322)进行生物信息学分析,了解其生物学特征,为下一步Nsp10的表达和ELISA检测方法的建立奠定基础。应用BioEdit、DNAStar、PsortⅡ、SignalP、TMHMM、ProtFun、Clustal_1.81、Mega等软件对Nsp10蛋白的理化参数、信号肽、跨膜区域、亲水性、表面可及性、抗原性、二级结构、亚细胞定位及与其他PRRSV毒株相应片段同源性等进行分析和预测。该蛋白理论分子质量为26.38 ku,pI为8.93,为稳定蛋白,不含信号肽,无跨膜结构,具有良好的形成抗原表位的条件。亚细胞定位结果表明,该蛋白极有可能位于细胞质,其编码氨基酸序列与参考毒株同源性为97.4%~100%。该蛋白保守性较高,是可溶性蛋白,有良好的抗原性,具备作为靶蛋白建立免疫方法的条件。

Abstract: The biocharacteristics of Nsp10 of HP-PRRSV were analyzed which in basis to express it and establish the ELISA method. BioEdit,DNAStar,PsortⅡ,SignalP,TMHMM,ProtFun,Clustal_1.81,Mega software packages were used to predict the physical and chemical properties,hydrophilicity,signal peptide,transmembrane,surface probability plot,antigenic index,secondary structure,subcellular localization and homology within the amino acid sequences of Nsp10. The predicted molecular weight was 26.38 ku and pI was 8.93. It was a stable protein and had no signal peptide and transmembrane. The study showed that Nsp10 possessed potential antigenicity and it mainly exerted the biological effects in the cytoplasm. The results showed that Nsp10 protein had highly homology with other PRRSV strain which selected. Nsp10 protein was a soluble protein with good conservatism and antigenicity. It can be a target protein used as coating antigen for ELISA detection.
