Effects of Dietary EPA/DHA Supplementation on Fermentation and Fatty Acid Profiles in Rumen Fluid from Artificial Rumen

The objectives of this study were to examine the changes of composition and content of fatty acid in ruminal fluid and the changes of volatile fatty acid in ruminal fluid when EPA/DHA was added in diet. The experiment using the RUSITEC(rumensimulation technique) apparatus. Treatments of the experimental diet were as follows: basal diet (CK, 40∶60 forage to concentrate),or a basal diet plus 0.65% EPA/DHA(Trt1) ,or basal diet plus 1.30%EPA/DHA(Trt2). Experimental period was consisted of 6 d for adaptation and 1 day for sampling. Culture fluid was collected every 2 h over a 12 h period on the last day of the experimental period. Compared with CK, C18∶0 decreased by 76.93% and 80.35% in Trt1 and Trt2, respectively; while the concentration of trans11C18∶1 increased by 185% and 127% in Trt1 and Trt2, respectively; and c9t11CLA increased by 147% and 222% in Trt1 and Trt2, respectively compared with CK. The concentration of VFA, transC18∶1 and c-9,t-11CLA in different sampling time were different. EPA/DHA supplementation could markedly affect VFA and fatty acids profile in rumen fluid.

The purpose of this experiment was to study the effect of different iron sources on the growth performance, immune function and iron status of weanling piglets. Ninety weanling piglets (Large White, 25 days of age) were randomly divided into 3 groups for three replicates of 10 piglets each (half male and half female). The piglets were fed with the diets containing 150 mg/kg ferrous sulfate, ferrous fumarate and ferrous glycine, respectively, for 28 d. No significant differences were observed on the growth performance during the trial (P>0.05). The diet with ferrous glycine increased IgG significantly compared with those with ferrous sulfate and ferrous fumarate (P<0.01) on 28 d. The diet with ferrous glycine increased RBC a little compared with that with ferrous sulfate (P=0.0518) on 14 d, while no difference was observed between ferrous fumarate and ferrous sulfate (P>0.05). The results showed that ferrous glycine is a better iron source than ferrous sulfate and ferrous fumarate for weanling piglets.

Four Aletai adult sheep with permanent cannula were chose, the total mixed ration (TMR) and the corn straw were normal feed every day , free drinking water. The different dry method were studied to the influence of dynamic degeneration rate in sheep rumen the various periods of duration Xinjiang small reed’s dry material, the crude protein ,the neutral detergent fiber (NDF) and the acid detergent fiber(ADF).The results showed that: ① The drying velocity is quick preservation of raw material nutrient were high, its degeneration rate is also high. Generally, natural dry is more than natural shade dry(P<0.05). ②In the early growing periods, the nutritional value of raw material is also more high, particularly the CP, the NDF and the ADF degeneration rate is quite high in the florescence and the solid time (P>0.05); ③Degradation rate of dry matter take 12 h as the turning point, The degeneration rat of crude protein arrives at 90% in four hour. Degradation rate of, the neutral detergent fiber (NDF) and the acid detergent fiber(ADF) , The increasing tendency relatively slow (P>0.05),there was no significant difference(except heading date).

A trial was carried out to study the effect of yeast culture on performance of lactating sows. The results indicate that yeast culture is able to increase the number of live littermate, weight of weaning pigs, rate of survival pigs during lactation etc. Furthermore, the results of this trial is to some extent similar to those of experiments carried out before. In conclusion, the results of this trial can be taken as practical guidance to improve efficiency of sow performance and profitability of swine farms. 

In the experiment 306 newlyhatched Avian broilers were conducted to study the effects of two kinds of phytase substitute inorganic phosphorus in diet on the growth performance, nutrient availability and excretion of feces P and N. There were 3 treatments with control group、experiment groupⅠ、experiment groupⅡ, and normal diet was fed to control group, lowP diet adding phytaseⅠwas fed to experiment groupⅠ, lowP with adding phytaseⅡwas fed to experiment groupⅡ. Each treatment had 3 replicates with 34 broilers per pen. The results indicated that ADG and ADFI of experiment group dietary supplementation with phytase were decreased(P>0.05); survival rates of broilers and the availability of Ca and P were increased significantly(P<0.05); the utilization of N was improved (P>0.05); and the excretion of N and P for broilers was decreased(P<0.05). the phytase Ⅰ was superior to phytase Ⅱ, diet added with phytase Ⅰ had significantly improved ADG and ADFI (P<0.05),and decreased the excretion of N (P<0.05),but excretion P had no significant between two groups.

We reported the application of AFLP markers to detect genetic polymorphic loci Xinji superfine wool sheep and their amplified APLP results. Their genetic construction of the population was also analyzed. Six APLP primers were used, genomic DNA from 24 sheep were restrictive by TaqⅠand EcoRⅠ,226 AFLP markers were obtained, the obtain marker numbers of individual primer were between 29 and 50. The results indicated as the following: AFLP marker was suitable for analysis genetic polymorphism in sheep, and the similarity index of population in Xinji superfine wool sheep was 0.914 to 0.982.The study provideed an useful parameter with appraise genetic stability of Xinji superfine wool sheep.

VP2 gene of infectious bursal disease virus(IBDV) was inserted into enhanced GFP expression vector pEGFP-C1. The recombinant eukaryotic expression vector pEGFP-VP2 was constructed, and detected by endonuclease digestion and PCR , and then was ransfected to COS7 cells by lipid reagent. After infecting, green fluorescence was observed under fluorescent microscope. The results indicates that GFP-VP2 fusion protein was successfully expressed in COS7 cells. Stable expressing cells was selected by 200 μg/ml G418, and GFP-VP2 fusion protain was purified by NiHis affinity column.

Neuromelanin (NM) is a complex polymer pigment found in catecholaminergic neurons of the human brain. It appears in greatest quantities in the human brain, in lesser amounts in some other nonhuman primates, but is absent from the brain in many lower species. Interest in this pigment has seen a resurgence in recent years because of a hypothesised link between neuromelanin and the especial vulnerability of neuromelanincontaining neurons to cell death in Parkinson’s disease (PD). 40 egg-laying chickens of 14-day-old were randomly divided into two groups, control and treatment. There were 20 chicks in each group. Newcastle vaccines (0.2ml/chick) were injected to cervical subcutaneous of control, and treatment was not only injected Newcastle vaccines, but also fed sustainedrelease compound immunopotentiator (1 particle/chick). Treatment were noticeably higher(P<0.01) than control in Newcastle disease (ND) HI antibody valence, rate of ERFC and positive ratio of ANAE after detected in 10, 20, 30 and 40 d; in addition, the growth of relative weight of the immune organs on treatment could be obviously promoted, compared with control, and ratio of chicken bursa and spleen also did, but the difference of them was not obvious(P>0.05).Therefore, the sustainedrelease compound immunopotentiator developed was an effective and less sideeffect immunopotentiator that had significant effect of immunoenhancement , disappeared slowly in delay phase ,and as well could promote the growth of organism.

The 18 spotted deers were studied, and they had been reported about antler production. According to antler production, high and low group were divided in the population. DNA isolated from each blood was used to amplify with PCR technique. All bands amplified with individual DNA was found in mixed DNA,which showed mixed DNA could be used to analyze the genetic structure of population in place of individual DNA. And compared with mixed DNA amplified bands of high and low group of each trait, there were 1 or 3 RAPD markertraits association in spotted deers population. These results indicated that tail analysis method could be used to detect QTL.

PCR-SSCP was used in this study to detect H-FABP 5’UTR polymorphisms in 16 Red Steppe hybrids crossed with Limousin. The correlationship was carried out between different genotype and meat quality.The results showed that 5’UTR of H-FABP gene exist polymorphisms in Red Steppe Crossed Herds, base T was inserted in 136 bp and the transformation of G→A in 142 bp. A genotype was negatively to the tenderness trait. The difference between AA with AB and BB was extremely significant (P<0.01) in tenderness, but AB and BB was no significant difference(P>0.05).The difference between genotype with the other meat quality was not significant (P>0.05). The effect was very significant of H-FABP gene to tenderness traits in Red Steppe Crossed Herds.

The buffalo oocytes were derived from abattoir ovaries , the effects of IVF of oocytes in different seasons were compared. The experiment has been lasted for two years, the data were divided into tree groups and statistically analyzed according to the oestrum cycle in buffalo. The results indicated that an average of 3.66 usable oocytes were derived from an ovary in oestrum season (Aug-Nov) which was significant higher than those in Mar-Jul and Dec-Feb (2.76 and 3.07 respectively, P<0.01). The cleavage rate and blastocyst rate of IVF oocytes obtained in different season showed no difference(P>0.05).

In order to study the effect of IL2 on the canine parvovirus vaccine efficacy, 40 puppies were randomly divided into control group and the experimental group, canine parvovirus vaccine and the vaccine plus 0.5 ml/head IL-2 injection was inoculated respectively. Post 7 days and 14 days, the standard test paper was used to detect antibody titer of the two groups. Results: The antibody titer revealed no significant difference between the test and control group before inoculation; Seven days post vaccination, antibody titer of the test group were very significant difference (P<0.01); 14 days post vaccination, there were very significant difference (P<0.01) between the test and the control groups in antibody titers. Experiments showed that IL-2 can significantly improve the efficacy of canine parvovirus vaccine. IL-2 is a good candidate canine parvovirus vauine adjuvant.

Chicken Newcastle disease virus(NDV) and infectious bronchitis virus(IBV)were diluted according to a certain proportion, then weve mixed equally, and the mixtures of the two virus were inoculated to 9~10 dog-old chicken embryos to research the proliferation of them in identical chicken embryos (abbreviating as homeomorphic cultivation). The results showed that: Inoculation of ND-Lasota strain (1∶5) and IBH120 strain (1∶5000) mixture, by allantoic cavity didnit influenced proliferation of the two virus in the identical chicken embryos. Hemagglutination titers of the two virus (NDV and IBV ) obtained after 96 h were respectively 11 log2 and 12 log2 by hemagglutination test, which were not lower than that of each one. Virus proliferation was tend to balance. Meanwhile, replication of NDV and IBV couldn’t produce interference effect each other under proper condition cultivation.

The effect of S boulardii on mice intestinal microflora imbalance was studied in this paper. Mice were treated with lincomycin hydrochloride to make microflora imbalance model. Then the models were treated by series dose of S. boulardii. The results showed that middle and low dose of S boulardii microcapsule had improved the intestinal microbial population disorder. The effective concentration of living cell is 1×108 to 5×108.

Materials about incidence of swine high fever syndrome were gained by epidemiological investigation of parts of Jinlin and Liaoning province. Porcine reproductive and respiratory syndrome virus, paramyxovirus virus and swine fever virus were mainly detected by electron microscope ; Serum testing swine high fever syndrome major pathogen antibody negative health piglets inoculated with paramyxovirus virus, virus solution, inactivated vaccine isolated from tissue samples in Jinlin province,meanwhile a negative control was made. Detection methods were used to study the clinical infection piglets including pathological testing, virus isolation, bacterial Detection. Pathogenic characteristics, morbidity regularity of swine high fever syndrome have been discussed as well as antibody dependent enhancement(ADE) which was taken place in the employment of inactivated vaccine. The results showed that: PRRSV was the major pathogen of swine high fever syndrome in the two provinces preliminarily and antibody dependent enhancement(ADE) phenomenon was concomitanted in the apply of inactivated vaccine.

This study is designed to analyze variation on body length and weight of crossbred between Germany Merino and Chinese Merino (Xinjiang type). Its body weight and body length changed. According to the analysis: the average weight of NO.1 crossbreed improved, average weight for NO.2 crossbreed increased more than its NO.1 crossbreed.The result show: NO.1 crossbreed are better than NO.2 crossbreed in Body length and body weight. So Germany Merino can improve the meat performance of Chinese Merino (Xinjiang) .

In this thesis, we studied the preparation of clove oil microcapsule by complex coacervation procedure using gelatin and gum arabic were used as wall material and clove oil for the core material, respectively. And the aim of the studies was mainly to approach the main effecting factors and suitable process. And the method of mensurating content of Eugenol was established. The results showed that the average particle size of clove oil microcapsule is 16 μm and it coating efficiency is 82.4%.Method of UV-vis spectrophotometry was established to determine content of Eugenol in the clove oil microcapsule. The standard curve equation is A=0.0171C +0.0014 (r=0.9994) in the concentration range of 1mg/ml to 10 μg/ml with its recovery rate of 98. 70%, and the method was extremely steady. Method of UVvis spectrophotometry established could be used to determine Eugenol in the clove oil microcapsule.
