Expression and Function Analysis of miR-143 in Muscle Cells

Abstract: This experiment was intended to study effects of microRNA-143 (miR-143) on muscle cell proliferation and differentiation of mice. Using the method of Real-time PCR to detect cell differentiation during the different time,and transfected miR-143 mimics and Inhibitor in C2C12 cells to test the expression of marker gene MyoG,then using the immunofluorescence and Western blotting method to test the MHC gene expression level.The results showed that miR-143 was closely related to the muscle cell proliferation and differentiation. Overexpression of miR-143,we identified that marker gene MyoG display a down-regulated expression,but after transfect siRNA,we found an opposite result. Also overexpression of miR-143 in differentiated cells,the result showed that expression of marker gene MHC was down-regulated. Therefore,miR-143 was a potential gene which had effects on muscle growth and development. For the future research,this study of muscle development and function could provide a new data.
