Establishment of SYBR Green Ⅰ Fluorescent Quantitative PCR Method for Detection of Porcine Reproductive and Respiratory Syndrome Virus

Abstract: A pair of specific primers conserved in the the Nsp7 genes of American porcine reproductive and respiratory syndrome virus (PRRSV) were synthesized. Optimization of reaction conditions, the detection of SYBR Green Ⅰ Real-time quantitative PCR method was compared and analyzed at the same time with the commercialization of fluorescent quantitative PCR kit. The results showed that the specificity of the method, high sensitivity, a good distinction between the American swine reproductive and respiratory syndrome virus and other viruses. 14 clinical samples were detected using this method, the positive rate was 85.7%, coincidence was 100% with commercial kits.
