Optimization and Analysis of In-gel Digestion with Bovine Spermatozoa Protein

Abstract: The achievemet ratio of identifying protein of sliver stained through MALDI-TOF-MS is low. The following variables were studied to effect of in-gel digestion of silver-stained 2-D gels with bovine spermatozoa protein :enzyme concentration, degestion time and dry gel. And optimization of in-gel digestion of silver-stained 2-D gels is including spot excision, destaing, digestion and extraction. Contaminating protein was analyzed and solution in this paper in order to enhance achievement ratio of protein identity
