Development of a LUX PCR Assay for Detection of Streptococcus suis Type 2

Abstract: The infection caused by Streptococcus suis type 2 is an important zoonosis. Based on the conserved cps2J region of Streptococcus suis type 2, the LUX primers were designed and the LUX PCR assay was developed to detect Streptococcus suis type 2. The sensitivity and specificity of the developed assay were analyzed by using the tonsils samples artificially contaminated with plasmid DNA as testing materials. Results showed that the developed Streptococcus suis serotype 2 specific LUX PCR assay was sensitive enough to detect 100 copies of plasmid DNA from the mask samples. The assay could distinguish Streptococcus suis serotype 2 from Streptococcus agalactiae, swine Pasteurellosis mutocida, Streptococcus faecalis, Streptococcus suis equi subsp equi, swine Cholerasuis and other swine bacteria pathogens. The developed assay can serve a candidate for the quick detection of Streptococcus suis serotype 2 infection in the field.
