Abstract: Interleukin-2(IL-2) gene was regarded as a candidate gene for immune function of Jingxinghuang chicken in this study. The genotype frequencies, allele frequencies, haplotypes and association with H/L of five SNPs were studied by direct sequencing. The results indicated that all the five loci fitted Hardy-Weinberg equilibrium and showed low degrees of heterozygosity with two genotypes detected.Meanwhile, the homozygosis genotype was predominant in all the five loci.Analysis of linkage disequilibrium (LD) suggested that SNP1 and SNP2 showed perfect LD, whereas SNP3, SNP4 and SNP5 showed perfect LD.Analysis of haplotypes revealed four haplotypes, among which Hap1 (TGACT) was predominant. The least square method (LSM) analysis showed that there were no significant difference between different genotypes and haplotype combinations(P> 0.05).The results suggested that more SNPs of IL-2 gene and association with additional immune traits would be investigated in future studies for better understand the roles in immune systems that chicken IL-2 playing.
