适度运动减轻流感小鼠肺脏炎性损伤的研究

Study on Moderate Exercise to Reduce Influenza Animal Lung Inflammatory Injury in Mice Model

摘要: 试验利用小鼠流感模型,研究适度运动能否减轻流感病毒引起的肺脏损伤,延长小鼠的生存期。将小鼠分为对照组和运动组,相同条件下饲养,对照组不进行踏轮试验,运动组小鼠进行踏轮试验(8~9 m/min,30 min/次,5次/周),第14天,经鼻腔接种致死量的鼠源性H1N1型流感病毒FM1,感染后第3天,对照组和试验组各处死5只小鼠,取其肺脏组织,各组分别取2个肺脏组织用于制作组织切片;余下的肺脏组织研磨后制备组织悬液用于检测病毒滴度和IL-6、TNF-α、MCP-1的含量。在感染病毒后3 d,运动组小鼠的肺脏炎症细胞浸润明显低于对照组,肺脏组织的IL-6、TNF-α、MCP-1的含量均显著低于对照组(P<0.05),肺脏组织的病毒滴度含量略低于对照组。此外,适度运动组的小鼠感染病毒后,生存期明显延长。结果表明,适度运动可显著降低流感病毒在小鼠肺脏引起的炎性损伤,明显延长小鼠的生存期。

Abstract: The purpose of this study was to explore whether moderate exercise could reduce inflammatory injury in lungs and prolong the survival of mice with influenza. The mice were divided into two groups (control and moderate exercise groups), control group’s mice were fed in the same condition as moderate exercise group, without treadmill exercise, the mice in moderate exercise group performed treadmill exercise (8 to 9 m/min, 30 min/d, 5 times/week for 2 weeks), all mice were nasally infected with FM1 influenza virus on day 14. On day 3 after being infected with the virus, five mice in exercise and control groups were killed for the lungs, two of which were for pathological analysis and three of which were for detection of IL-6, TNF-α, MCP-1 and virus titer. On day 3 after infected with the virus, there were lower inflammatory cells, virus titers and IL-6, TNF-α, MCP-1 (P<0.05) in the lungs of mice in exercise group than that in control group, furthermore the survivals of mice in exercise group were also apparently longer than that in control group. Moderate exercise could significantly reduce the inflammatory injury in lungs of the mice with influenza and apparently prolong the survival of the mice.
