Cloning, Sequence Analysis and Expression of Lbx2 Gene in Pig

摘要: 试验利用PCR扩增猪Lbx2基因组序列,通过克隆测序寻找基因中的突变位点,采用半定量RT-PCR分析猪不同组织中Lbx2基因的表达情况。结果显示,Lbx2基因在肝脏、肾脏和脾脏中微弱表达,在肌肉组织中不表达;通过比较不同物种氨基酸序列,结果发现猪Lbx2与牛、猩猩、犬进化关系较近;此外,在猪Lbx2基因组中发现5处突变位点,其中1个缺失突变(缺失11 bp)位于基因启动子区,另外4个单核苷酸突变分别位于第1外显子和内含子中。该试验结果为下一步Lbx2基因的功能研究奠定了基础。

Abstract: In this study, the Lbx2 gene was amplified by PCR, then the gene mutation was identified by sequencing. The gene expression profile of different tissues in pig was analyzed by semi-quantitative RT-PCR method. The results showed that Lbx2 was lowly expressed in liver, kidney and spleen. A phylogenetic tree was constructed by aligning the amino acid sequences of different species, which revealed that the porcine Lbx2 had a closer genetic relationship with the Lbx2 of Bos taurus, Pongo abelii, Canis familiaris than with those of other species. Moreover, five mutations were detected in the Lbx2 genomic fragment, including one deletion mutation and four single nucleotide polymorphisms. These results would provide a foundation for further studies on Lbx2 function.
