复方中药制剂对H22荷瘤小鼠免疫功能的影响

Effect of Compound Chinese Medicine Preparations on Immune Function of H22 Tumor-bearing Mice

摘要: 【目的】探究蛹虫草、灵芝、黄芪、五味子、党参、当归、茯苓复方中药制剂对H22荷瘤小鼠免疫功能的影响。【方法】建立H22荷瘤小鼠模型,随机将小鼠分为模型组、复方中药制剂高(250 mg/(kg·d))、中(50 mg/(kg·d))和低(10 mg/(kg·d))剂量组,各剂量组灌胃给药,1次/d,模型组灌胃同等体积生理盐水,试验期3周,另设置正常组进行常规饲喂。末次给药后采集抗凝全血,检测小鼠血细胞数量、血清中超氧化物歧化酶(SOD)活性以及丙二醛(MDA)、细胞因子水平。用流式细胞术检测外周血CD4+/CD8+ T细胞亚群;通过小鼠脾淋巴细胞转化试验、碳粒廓清试验考察荷瘤小鼠免疫活性。【结果】与正常组相比,模型组小鼠血液中白细胞(WBC)、血小板(PLT)、淋巴细胞(W-SCR)数量极显著或显著减少(P<0.01;P<0.05),白细胞介素-2(IL-2)、IL-6和肿瘤坏死因子-α(TNF-α)含量、SOD、半胱氨酸蛋白酶3(Caspase-3)活性以及细胞色素C(Cyt C)浓度均极显著或显著降低(P<0.01;P<0.05)。与模型组相比,高、中和低剂量组小鼠血清IL-2、IL-6、TNF-α含量和SOD、Caspase-3活性均极显著或显著升高(P<0.01;P<0.05),MDA含量均极显著或显著降低(P<0.01;P<0.05);中剂量组小鼠血液中WBC、PLT数量极显著或显著增加(P<0.01;P<0.05);高剂量组小鼠血液中PLT数量极显著增加(P<0.01),Cyt C浓度显著升高(P<0.05)。流式细胞检测结果显示,模型组小鼠血液T淋巴细胞CD4+、CD8+亚群细胞数量较正常组显著减少(P<0.05)。与模型组相比,复方中药制剂高、中剂量组小鼠血液T淋巴细胞CD4+、CD8+亚群细胞数量显著增加(P<0.05),高、中剂量组小鼠刺激指数、半数溶血值和吞噬指数均极显著或显著升高(P<0.01;P<0.05)。【结论】复方中药制剂能提高H22荷瘤小鼠免疫活性,进而促进肝癌细胞凋亡。

Abstract: 【Objective】 The experiment was to investigate the immunoregulatory effect of the compound Chinese mdicine preparations extracted from Cordyceps militaris,Gantinoderma lucidum,Astragalus membranaceus,Schisandra chinensis,Codonopsis pilosula,Angelica sinensis and Poria Cocos on H22 tumor-bearing mice.【Method】 The model of H22 tumor-bearing was established,and the mice were randomly divided into model group,high (250 mg/(kg·d)),medium (50 mg/(kg·d)) and low (10 mg/(kg·d)) dose groups of compound Chinese medicine preparation,each dose group was administrated intragastric once a day,and the model group was administrated intragastric with the same volume of normal saline.The experiment period was 3 weeks.The normal group was also set up for routine feeding.Whole anticoagulant blood was collected after the last administration,and the number of blood cells,the activity of superoxide dismutase (SOD) and the levels of malondialdehyde (MDA) and cytokines in serum were detected.Peripheral blood CD4+/CD8+ T cell subsets were detected by flow cytometry.The immune activity of tumor-bearing mice was investigated by spleen lymphocyte transformation test and carbon clearance test.【Result】 Compared with normal group,the number of white blood cells (WBC),platelets (PLT) and lymphocytes (W-SCR) in the blood of the model group was extremely significantly or significantly decreased (P<0.01 or P<0.05),the contents of interleukin-2 (IL-2),IL-6 and TNF-α,activities of SOD,Caspase-3 and concentration of cytochrome C (Cyt C) were extremely significantly or significantly decreased (P<0.01 or P<0.05).Compared with model group,the contents of IL-2,IL-6 and TNF-α in serum and the activities of SOD and Caspase-3 in high,medium and low dose groups were extremely significantly or significantly increased (P < 0.01 or P < 0.05),MDA content was extremely significantly or significantly decreased (P<0.01 or P<0.05).The amount of WBC and PLT in the medium dose group was extremely significantly or significantly increased (P<0.01 or P<0.05).The number of PLT in high dose group was extremely significantly increased (P<0.01), and the concentration of Cyt C was significantly increased (P<0.05).Flow cytometry showed that the number of T lymphocyte CD4+ and CD8+ subsets in model group was significantly decreased compared with that in normal group (P<0.05).Compared with model group,the number of T lymphocyte CD4+ and CD8+ subsets in high and medium dose groups was significantly increased (P<0.05),and the stimulation index,median hemolysis value and phagocytosis index of mice in high and medium dose groups were extremely significantly or significantly increased (P<0.01 or P<0.05).【Conclusion】 The compound Chinese medicine preparation could improve the immune activity of H22 tumor-bearing mice,and then promote the apoptosis of liver cancer cells.
