Evaluation on the Genetic Diversity of Guizhou Pony and Ili Horse Using Microsatellite Markers

Abstract: Nine microsatellite loci were chosen to detect the genetic diversity of Guihzou pony and Ili horse in this experiment. Polymerase chain reaction (PCR) products were detected by 8% native polyacrylamide gel electrophoresis in silver staining protocol. Allele frequency numbers of effective alleles (Ne),heterozygosity (H) and polymorphism information contents (PIC) for each microsatellite locus were calculated by POPGENE software. The greatest variation was found from COR059, while the lowest was from ATH040 in all of 9 microsatellite loci. Mean numbers of Ne, H and PIC in Ili horse were higher than that in Guizhou pony. Mean coefficient of gene differentiation (GST) of whole breeds was 0.041. It suggested that the gene of Guizhou pony was more homozygous than that of Ili horse, and both of them showed plenty of genetic variation.
