Genetic Diversity among Five Black Goat Breeds(Populations) Based on Microsatellite Analysis

Abstract: Genetic diversity for Pangzhihua Black goat (PZH) and other four black goat breeds(populations),including Janchang Black goat (JC),Yunling Black goat (YL),Jintang Black goat (JT), Lezhi Black goat (LZ). We reestimated using fourteen microsatellite makers by PCR, polyacrylamide gel electrophoresis, and silver staining of modified Sanguintti method. The results showed that all of the microsatellites were highly polymorphic loci,the polymorphic information content (PIC) varied from 0.5129 to 0.7646. Each of the five breeds(populations) possesses had a high level of genetic variability, the PIC and the mean heterozygosity (H) varied from 0.6405 to 0.6856 and 0.6985 to 0.7371,respectively. But the Wright’s F-statistics of subpopulation within total (Fst) showed a low mean value 0.0296,suggesting the variation between populations was only 2.96% share and gene flow was exist among groups. Nei’s genetic distances (D) were calculated, which varied from 0.0365 (between JT and LZ) to 0.1286(between PZH and JT). In the NJ cluster analysis, PZH, JC and YL could be separated to one cluster, JT and LZ to the other. Additionally, the result displayed a remarkable degree of consistency with their different geographical origins.
