The Impact of Tautrine on SOD,GSH-PX,MAD and T-AOC in Extracellular Fluid of STZ-induced Islet Cell

Abstract: To approach the protective effects of taurine on islet cells treated with streptozotocin,then provide a theoretical basis for the treatment of diabetes.250 to 300 g rats were in situ perfusion with collagenase V in bile common duct after anesthesia. Pancreatic tissues were digested using aqueous bath under 38 centigrade. Islet cells were purified by Histopaque 1077 density gradient centrifugation,and the purity was detected by DTZ,the cytoactive was detected by glucose stimulation. Purified islet cells were randomly divided into 6 groups,normal control group (control group),taurine control group,streptozotocin group(STZ group),high-dose taurine group (20 mmol/L Tau group),medium-dose taurine group (10 mmol/L Tau group) and low-doses taurine group (5 mmol/L Tau group). The cells were cultured for 24 h,then the level of superoxide dismutase (SOD),glutathione peroxidase (GSH-Px),malondialdehyde (MDA),total antioxidant capacity (T-AOC),were measured.The results showed that the three doses of taurin groups and taurine control group could significantly increase the levels of SOD,GSH-Px,T-AOC in extracellular fluid compared with STZ group,and could significantly decrease the level of MDA compared with STZ group.Therefore,taurine could protect and repair islet cells by way of increasing the oxidation resistant ability of islet cells induced with STZ.
