The Primary Culture of Fin Cells from Carp

Abstract: Through the primary culture of the carp fins cells,infered the optimum culture conditions. The major experiment method is tissue explant technique. A small piece of fins were cultivated in different conditions(media, temperature, pH, CO2), the growth status were observed. The cells migrated out around tissues after 48 hours and the growth halo emerged. Under this condition, monolayers of primarily cultured renal cells were routinely obtained within a week,most of the cultured cells were epithelioid macrophages and little into fiber appearance cells. The optimized conditions for carp fin cells were: medium M199, 25 to 27 ℃, pH 7.2 to 7.6, the concentration of CO2 was 5%, the concentration of serum for primary culture was 15% to 20%, the purification fin cells could be getten after 12 days.
