猪β2m在pMAL-p2X系统中的表达与纯化

Abstract

Abstract: Swine β2m was optimized to express in pMAL-p2X from several aspects including different pH value of the medium,induction temperature (T),cell density,concentration of IPTG and induction time (t),under the optimum expression condition,swine β2m was purified under amylose affinity resin column. The results showed that the optimum expression condition for β2m in pMAL-p2X was pH=7.5,T=37 ℃,D₆₀₀_nm =0.3,IPTG=0.5 mmol/L,and t=5 h. Under the optimum expression condition, the relative expressed content of recombinant β2m protein reached to 45%, the purity of β2m protein was more than 90%.
