Sex Identification in Goat by Amplification of Amelogenin (AMEL) Gene

Abstract: Based on the characteristic of the hircine AMEL-X and AMEL-Y gene with various degree deletions,a pair of primers were utilized to establish the process of sex identification by PCR and agarose gel electrophoresis with mixed samples of the hircine blood genomic DNA, and the PCR produced was purified, cloned and sequenced, then sex identification to the 46 hircine blood samples(♀:24,♂:22).The results showed that the male hircine sample produces a non-specificity band and a specificity band, while the female hircine sample produces one band. The non-specificity fragment 349 bp, the specificity fragment 289 bp. By comparison, the results of the 46 hircine blood DNA samples sex identification with the hircine actual sex, the female accuracy 100%(22/22), the male accuracy 100%(24/24).
