Cloning and Sequence Analysis of Mitochondrial Cytochrome c Oxidase Subunit 1 of Oesophagostomum quadrispinulatum Isolates from Changsha City

Abstract: To analyze the differences in cytochrome c oxidase subunit 1 (cox1) gene of Oesophagostomum quadrispinulatum isolated from Changsha city. The partial cox1 (pcox1) was amplified from each O.quadrispinulatum sample,and pcox1 sequences were aligned using the Clustal X 1.83. Sequence homology analysis was performed using the MegAlign program of the software DNAStar version 5.0 by comparing with the corresponding sequence of O.quadrispinulatum available in GenBankTM. The results showed that the lengths of pcox1 were 393 bp. Sequence comparison revealed that the similarity in pcox1 sequences among Changsha isolates and that of the O.quadrispinulatum available in GenBankTMwere over 98%. There was no significant variation in pcox1 sequences within O.quadrispinulatum,while inter-species difference was obviously.The results provided a foundation for further studies of molecular identification and molecular genetics of O.quadrispinulatum.
