The Effect of Raffinose on Frozen/Thawed Ujumqin Sheep Semen Quality

Abstract: The aim of the present study was to determine the effects of different doses of raffinose on post-thawed sheep spermatozoa quality. Ejaculates collected from 10 healthy Ujumqin sheeps were evaluated and pooled at 37 ℃. Semen samples were divided equally into 5 groups, then diluted with 0, 5, 10, 15, 20 mmol/L of different concentrations of raffinose and frozen in 0.25 mL French straws. Frozen straws were thawed individually at 37 ℃ for 15 s in a water bath for microscopic evaluation and then detected the activity of SOD, GSH, MDA, GSH-Px and CAT. The results showed that, compared to the control groups, when the addition of raffinose was 15 mmol/L, sperm motilities were increased significantly (P<0.05), abnormal sperm rate was decreased significantly (P<0.05), host rate was increased significantly (P<0.05), the enzyme activity of SOD significantly improved (P<0.05), MDA was decreased significantly (P<0.05).
