Influence of Buffalo PBMC Activity by Different Resuspension Methods During Washing Process

Abstract: The peripheral blood mononuclear cell (PMBC) activity of buffalo was comported by different resuspension methods during washing process.The PBMC was separated by density gradient centrifugation,then cell was resuspended by transferpettor-spirting method & centrifuge tube-sloshing method, when cell was washed three times the activity was evaluated. The cell activity by transferpettor-spirting method & centrifuge tube-sloshing method was(74.9±2.6)% and(98.5±0.5)% respectively, the difference of cell activity was significant(P<0.01). The activity of PBMC was higher by centrifuge tube-sloshing, which could provide an useful method for further study that requiring higher cell activity. 
