蛋鸡骨骼肌卫星细胞分离培养及鉴定

摘要: 采用组织块分离法从蛋鸡鸡胚腿部肌肉组织中体外培养获取鸡骨骼肌卫星细胞,对培养的细胞进行了传代、冻存和复苏研究,同时从分子表达水平对分离培养的卫星细胞进行了鉴定,建立了一套鸡骨骼肌卫星细胞分离、传代、冻存和复苏的方法。特异基因表达鉴定结果表明,该方法分离得到的细胞能够表达卫星细胞特异的标志基因Pax7,而肌细胞分化基因MyoD、Myogenin未表达,说明获得的肌卫星细胞纯度较高,尚未进行肌细胞的分化。

Abstract: Chicken skeletal muscle satellite cells were obtained from the leg muscle tissue of chicken embryo by tissue separation. The cells were studied on passage,cryopreservation and resuscitation,and identified by specific gene expression assays, and a new method was established on isolation,passage cryopreservation and resuscitation of chicken skeletal muscle satellite cells. The results of specific gene expression showed that the cells could express specific marker genes Pax7,but not MyoD,Myogenin of the myogenesis genes of muscle satellite cell,which indicated that the satellite cells were of higher purity and didn’t differentiate into skeletal muscle cell.
