鸭疫里氏杆菌PCR快速检测方法的建立

Establishment of PCR Method for Quick Detection of Riemerella anatipestifer

摘要: 本试验根据GenBank中的鸭疫里氏杆菌主要外膜蛋白A基因序列,在其保守区设计了1对引物,建立了检测鸭疫里氏杆菌的PCR方法,对鸭疫里氏杆菌进行检测,结果能扩增出预期片段,而金黄色葡萄球菌、链球菌、多杀性巴氏杆菌、沙门氏菌、大肠杆菌的扩增结果均为阴性。说明该PCR法具有较好的特异性,可用于鸭疫里氏杆菌感染的检测、分子流行病学调查和分离菌株的快速鉴定。

Abstract: According to the gene sequences of the outer membrane protein A from Riemerella anatipestifer(RA) in GenBank, one pair of primers were selected and a specific PCR assay was made. Riemerella anatipestifer were found positive in the PCR. Staphyloccocus aureus, Rosenbach, Streptococcus, Pasteurella Multocida, Salmonella and Escherichia coli were all found negative in the PCR. The specificity and reproduction of PCR suggest it can be used for routine identification of Riemerella anatipestifer.
