Study on Extraction and Purification of Baicalin from Hairy Roots of Scutellaria baicalensis Georgi

Abstract: Using the content of Baicalin as the standard, the optimum macroporous resin was selected, and the factors of Baicalin's extraction and purification were investigated. LX-68 macroporous resin was the suitable one, Baicalin was extracted under the optimum conditions of a sample concentration of 8.69 g/L, a adsorption speed of 6 BV/h, a sample volume of 16 BV, a diameter-high ratio of 1:14, a elution reagent of 70% alcohol, a elution speed of 6 BV/h, a elution volume of 10 BV, and then purificated by silica gel column chromatography, the condition was chloroform-ethyl acetate-methanol-formic acid(7:3:1.5:0.5). The LX-68 macroporous resin could be used to extract Baicalin from the hairy roots of Scutellaria baicalensis Georgi, the adsorption capacity of total flavone was 131.2 mg/g, the returns-ratio was above 69.1%, and the purity of Baicalin was 98.45%.
