Cloning and Sequence Analysis of Microneme Proteins 3 Gene of Different Eimeria tellena Strains

Abstract: The gene coding for Eimeria tenella microneme protein 3 (EtMIC3) of different strains was amplified by RT-PCR, subsequently cloned and analysed, and compared with related sequences published on GenBank. The results showed that the EtMIC3 sequence of 8 different strains was 2976 bp in length, with little difference in each other. The sequence similarities of nucleotide were 99.8% to 99.9% compared with FJ374765.1 CDs available in GenBank, and 99.9% to 100.0% within each of EtMIC3 obtained in this study with no obvious difference between strains of different geographical origin or drug resistance. The sequence similarities of induced amino acid were 99.6% to 99.9% compared with EtMIC3 (ACJ11219) available in GenBank. It was the first report on the analysis of EtMIC3 sequences of different Eimeria tenella strains from China. The results revealed EtMIC3 was high conservative, and could be used as valid vaccine candidate for the further study on constructing genetic engineering vaccine.
