Studies on Single Nucleotide Polymorphisms at the Exon 2 of H-FABP Gene among Different Sheep Breeds

Abstract: PCR-SSCP approach was applied to test the single nucleotide polymorphisms (SNPs) at the exon 2 of heart fatty acid-binding protein (H-FABP) gene and four sheep populations that comprised of Northeast fine wool sheep, Small-Tailed Han sheep, Dorper sheep and Dorsets sheep were screened in this study. The results showed that four SNPs at 939(A/G), 980(G/A), 1013(A/C), 1018(T/C) and three genotypes were detected. During the Northeast fine wool sheep, Small-Tailed Han sheep and Dorper populations, the BB was the predominant genotype. While in Dorsets sheep, the AB was the predominant genotype. A chi-square analysis suggested that the allele frequencies and genotype frequencies of H-FABP gene were in Hardy-Weinberg equilibrium. Statistical analysis revealed an intermediate polymorphism information content (PIC) in the Northeast fine wool sheep, Dorper sheep and Dorsets populations (0.25<PIC<0.50), but a low PIC in the Small-Tailed Han sheep population (PIC<0.25). Further analysis showed that the different groups had different degree of genetic variation.
